Objective:Detection the expression of PIM-1 in androgen-dependent,androgen deprivation therapy and castrate-resistent prostate cancer prostate cancer tissues,and explore the clinical significance of PIM-1 inhibitor Quercetagetin(Qg)in the process of androgen-dependent to castrate-resistant prostate cancer.Methods:Immunohistochemical,RT-PCR,Western Blot method was used to detect the protein expression of PIM-1 in androgen-dependent,androgen deprivation therapy and castrate-resistent prostate cancer tissues.Statistical method was used to analysis the PIM-1 expression differences in three tumor tissues.Using established androgen-dependent and castrate-resistent prostate cancer tumor model in nude mice using LNCaP cells,draw the growth curve by using PIM-1 inhibitor Quercetagetin(Qg),Finding different expression of PIM-1 in different tumor tissues.Results:1.Human tissues immunohistochemistry:1)human castrate-resistant prostate cancer tissues PIM-1 protein expression was higher than androgen-dependent tissues.And there was significant difference(P<0.01).2)human castrate-resistant prostate cancer tissues PIM-1 protein expression was higher than androgen deprivation therapy prostate cancer tissues.And there was no significant difference(P>0.05).2.RT-PCRl)human castrate-resistant prostate cancer tissues PIM-1 protein expression was higher than androgen-dependent tissues.And there was significant difference(P<0.01).2)human castrate-resistant prostate cancer tissues PIM-1 protein expression was higher than androgen deprivation therapy prostate cancer tissues.And there was no significant difference(P>0.05).3.Western Blot:1)human castrate-resistant prostate cancer tissues PIM-1 protein expression was higher than androgen-dependent tissues.And there was significant difference(P<0.01).2)human castrate-resistant prostate cancer tissues PIM-1 protein expression was higher than androgen deprivation therapy prostate cancer tissues.And there was no significant difference(P>0.05).4.Growth curve and PIM-1 inhibitor Quercetagetin(Qg),:1-4th groupe stands for Quercetagetin+castration,Quercetagetin+uncastration,control+castration,control+unc-astration respectively.1th groupe and 2th groupe tumor tissue grow slower than 3th groupe tissues.And there was significant difference(P<0.05).3th groupe tumor tissue grow slower than 4th groupe tissues.And there was no significant difference(P>0.05).5.IHC of PIM-1 in different tumor tissues:PIM-1 inhibitor Quercetagetin inhibite PIM-1 protein expression.Conclusions:1.PIM-1 is overexpressed in castrate-resistant prostate cancer tissues.PIM-1 plays an important role in the castrate-resistant prostate cancer transformation process,which may be related to the role of androgen receptor signaling pathway or other signaling pathway.2.PIM-1 inhibitor Quercetagetin(Qg)inhibite PIM-1 protein expression and growth in tumor of nude mice model,Qg has some clinical significance for the treatment of prostate cancer and open up new prostate cancer therapeutic areas especially for castrate-resistant prostate cancer.
