| Objective: Stomach cancer which is a global malignancy,own a high incidence and mortality rates.micro RNA exists widespread in eukaryotic cells,which was involved in many physiological processes such as proliferation,differentiation,apoptosis and self-renewal of stem cell,and playing an important role in the occurrence,development and metastasis of tumor.Studies had found that the expression of miR-191 and miR-135 b in gastric cancer tissue was higher than that in normal population.This paper was to look for effective molecular markers of gastric cancer by researching the expression of miR-191-5p and miR-135b-5p in peripheral blood so that the gastric cancer can be screened better..Methods: 1.Clinical and pathological data of 110 cases were selected from the satisfactory individuals,of which 35 cases were classified into control group while 75 cases attaching by gastric cancer were classified as the experimental ones.The experimental cases were diagnosed with gastric cancer according to gastroscope pathological diagnosis and without surgery or chemotherapy.2.Peripheral blood samples of the 110 cases were acquired followed by the procedure of preconditioning,namely obtaining the supernate by centrifugating the peripheral blood samples in a tube containing separation gel.3.miRNA was extracted and enriched,91 eligible cases were chosen for the next experiment.4.The level of expression of miR-191-5p and miR-135b-5p were tested in serum by RT-PCR method.5.All results were determined by the statistical analysis.Conclusion : miR-191-5p and miR-135b-5p expressed in peripheral blood of both individuals attached with or without gastric carcinoids,however,the concetration of miR-191-5p and miR-135b-5p in experimental cases were obviously higher than that incontrols. |
