Objective:To investigate the effects of metformin,full-length adiponectin(f-Ad)and globular adiponectin(g-Ad)on proliferation of human colon cancer SW480 cell line cultured in vitro and to elucidate its possible mechanism and meaning.Method:In vitro culture of Human Colorectal Cancer Cell SW480,After treated with different concentration of metformin(5mmol/L?10mmol/L?20mmol/L),f-Ad and g-Ad(5?g/ml?10?g/ml?20?g/ml),the cell viability of all groups was measured by CCK-8 assay.After treated with 20mmol/L metformin,20?g/ml f-Ad and 20?g/ml g-Ad for 48 hours.The relative expression level of STAT3(Signal transducer and activator of transcription 3)and Survivin protein in control group and intervention group were detected by Western Blot assay.Result:1.The CCK-8 assay showed that compared with the control group,the intervention group with different concentrations f-Ad and g-Ad(5?g/ml?10?g/ml?20?g/ml),metformin(5 mmol / L,10 mmol / L,20 mmol / L)for 48 h were inhibited.With the increase of drug concentration,the inhibition rates of proliferation present a dose-dependent manner.The inhibition effect of merformin is the most obvious,g-Ad is second,f-Ad is the last.2.Western Blot analysis showed that compared with the control group,the relative expression level of STAT3 protein in metformin(20mmol/L)and g-Ad(20?g/ml)intervented groups are decreased(P >0.05),but f-Ad(20?g/ml)intervented group is decreased(P <0.05).The relative expression level of Survivin protein in metformin(20mmol/L),g-Ad(20?g/ml)and f-Ad(20?g/ml)are all significantly decreased(P <0.05).Conclusion: 1.Metformin,g-Ad and f-Ad inhibit the growth of colon cancer SW480 cell line,for a dose-dependent manner.2.The growth inhibitory effect on colon cancer SW480 cell line which is mainly through inhibiting cell proliferation and reducing the expression of apoptosis inhibiting protein.3.F-Ad inhibits the expression of Survivin protein,metformin and g-Ad inhibit the expression of Survivin and STAT3 protein,so metformin and g-Ad inhibit the cell growth through STAT3/Survivin pathway.Objective:Analysis the relationship between the Mets(metabolic syndrome)related indicators and the level of serum APN(adiponectin)on the colorectal cancer patients.Analysis the relationship between the metabolic syndrome related indicators and the level of cancer tissue adiponectin m RNA expression on the colorectal cancer patients.To investigate the relationship and meaning between the adiponectin and the metabolic syndrome on colorectal cancer patients.Method:Collected 80 colorectal cancer cases which selected by research group,to analysis the relationship of the WC(waistline circumference),BMI(body mass index),BP(blood pressure),FPG(fasting plasma glucose),serum adiponectin,HDL-C(high density lipoprotein),TG(triglyceride)and other indicators and the expression of adiponectin in tumor tissue detected by RT-PCR(real-time fluorescent quantitative reverse transcription polymerase chain reaction)method.Divided these patients by the level of serum adiponectin and the expression of adiponectin in tumor tissue from low to high into 4 groups(Q1-Q4),with 20 cases in each group.Divided these patients by whether suffer the metabolic syndrome or not into 2 groups(metabolic syndrome 17 cases,non metabolic syndrome 63 cases).Compared the WC,BP,FPG,HDL-C,TG,Hb A1C(glycosylated hemoglobin),HOMAIR(homeostasis model assessment of insulin resistance),APN in each group with Oneway ANOVA method.Analysis the relationship between the levels of adiponectin m RNA expression and other indicators with Pearson correlation analysis method.Result:1.In the colorectal cancer patients with the metabolic syndrome,abnormal WC and BP were in the majority and the majority of patients has 1 ~ 2 abnormal indexes,males with abnormal number are more than females,which TG and HDL-C abnormalities of men significantly more than women.2.With the increase of serum APN level,BMI and WC was decreased.Compared with Q4,the differences of Q1,Q2,Q3 were statistically significant(P < 0.05);The FBG,HOMA-IR,Hb A1 c was also decreased,the difference between Q2 and Q4 was statistically significant(P < 0.05);the serum TG,HDL-C and DBP,SBP has no statistical difference between the four groups(P > 0.05).3.With the increase of the level of APN in cancer tissues,WC,SBP(systolic blood pressure),DBP(diastolic blood pressure),HOMA-IR showed an increasing trend,only SBP in Q1 and Q2 group had significant difference(P < 0.05).Other indexes between different groups were not statistically significant(P > 0.05);FBG,Hb A1 C showed a downward trend,but the differences among the four groups with no statistical significance(P > 0.05),TG,HDL-C,BMI changing tendency is not obvious,and the differences between the four groups of no statistical significance(P > 0.05).4.The serum adiponectin and HDL-C in metabolic syndrome group was significantly lower than them in non metabolic syndrome group(P < 0.001),The WC and HOMA-IR in metabolic syndrome group was significantly higher than them in non metabolic syndrome group(P < 0.001),BMI was higher than non metabolic syndrome group(P < 0.01),FBG and Hb A1 C was higher than non metabolic syndrome group(P< 0.05),while BP,TG and cancer tissue adiponectin m RNA between two groups were not statistically significant(P < 0.05).5.There was no correlation among the level of cancer tissue adiponectin m RNA expression and age,BP,BMI,WC,FBG,HOMA-IR,Hb A1 C,blood lipid(P >0.05).Conclusion:1.In colorectal cancer patients,the different levels of serum adiponectin has the different metabolic syndrome related indicators,with the elevation of serum adiponectin,the FBG,HOMA-IR,Hb A1 C,BMI and WC depressed,TG,HDL-C,BP had no obvious changed.2.In colorectal cancer patients,there was no correlation among the level of cancer tissue adiponectin m RNA expression and age,BP,BMI,WC,FBG,HOMA-IR,Hb A1 C,blood lipid(P>0.05).3.The colorectal cancer patients with metabolic syndrome has lower serum adiponectin than without metabolic syndrome,and the level of cancer tissue adiponectin m RNA expression in both groups had no significant difference.
