Objective: To investigate the expression of PD-1 and PD-L1 in nasopharyngeal carcinoma and their relationship with the clinicopathological characteristics and prognosis of patients with nasopharyngeal carcinoma?Methods: 1. Formalin-fixed, paraffin-embedded tissue by nasopharyngeal biopsy from 120 patients with histological diagnosis of nasopharyngeal non keratinizing carcinoma were collected? 2. By immunohistochemistry staining, the expression of PD-1 and PD-L1 in nasopharyngeal carcinoma tissue were detected. 3.To collect the clinical pathological features of nasopharyngeal carcinoma patients, including gender, age, smoking history, pathological type, stage and lymph node metastasis. According to American Joint Committee on Cancer(AJCC) staging in nasopharyngeal carcinoma TNM staging system(2010 Edition). 4.Patients with nasopharyngeal carcinoma were followed up for disease free survival(DFS). 5.Using SPSS 16.0 statistical software to analyze the data. Chi-square test was used to assess the expression of PD-1 and PD-L1 correlated with clinical pathological characteristics. Survival analysis was depicted by Kaplan–Meier method.Results: 1.The positive rate of PD-1 expression in nasopharyngeal carcinoma tissue was 29.2%(35/120), and the positive rate of PD-L1 expression was 70.8%(85/120). 2.The expression of PD-1 in nasopharyngeal carcinoma tissue was not related to age, sex, pathological type, stage and lymph node metastasis(P > 0.05), but related to smoking(P=0.026).3.The expression of PD-L1 in nasopharyngeal carcinoma was not related to gender, smoking, pathological type and lymph node metastasis(P > 0.05), but related to age(P=0.006) and stage(P=0.036). 4. After conventional chemoradiotherapy, There was no significant difference in disease-free survival(P=0.191) in nasopharyngeal carcinoma patients with PD-1 negative and PD-1 positive.Compared with PD-L1 low expression, The nasopharyngeal carcinoma patients with PD-L1 high expression showed poor disease free survival(P=0.002).Conclusion: 1.High expression of PD-L1 in nasopharyngeal carcinoma tissue can significantly shorten disease-free survival of patients with nasopharyngeal carcinoma after conventional chemoradiotherapy to receive complete response, but the expression of PD-1 not. 2.The expression of PD-1 in nasopharyngeal carcinoma tissue was not related to age, gender, pathological type, stage and lymph node metastasis, but related to smoking. 3.The expression of PD-L1 in nasopharyngeal carcinoma tissue was not related to gender, smoking,pathological type and lymph node metastasis, but related to age and stage.
