The Expressions Of E-cadherina And Ki-67 In Gastric Cancer And Their Significance

Objective: In the research of all kinds of malignant tumors in our country, gastric cancer occurrence and development is still the focus. In this article, we use immunohistochemical method to detect E-cadherin and Ki-67 expression in gastric cancer tissue to study its correlation with gastric cancer process, in order to further discuss the molecular mechanism of the development and metastasis of gastric cancer, and provide theoretical basis for gene targeting treatment of gastric cancer.Methods:Collect concurrent pathologic diagnosed gastric cancer surgery resection specimen of 62 cases from the fourth affiliated hospital of Nanchang University from January 2011 to December 2013. Gastric cancer specimens are divided into several groups, including gender group, age group, tumor diameter group, tumor site group, degree of differentiation, infiltration depth, lymph node metastasis, distant metastasis, TNM staging, using immunohistochemical method(Max Vision) in the detection of gastric cancer tissue samples of E cadherin and positive rate and negative rate of Ki-67 expression and also normal gastric tissue, and then use SPSS22.0 to analyze the correlation between them and between groups and undertake statistical analysis of experimental data, and the relationship between E cadherin and Ki-67.Results:(1)in 62 cases of gastric cancer tissues, the positive rate of E-cadherin and Ki-67 is 48.4%(30/62) and 61.3%(38/62) respectively.(2)E- cadherin in normal gastric tissues were 100%, higher than expression in gastric cancer tissue, statistically significant difference(P < 0.05). E-cadherin in high and middle differentiation in gastric cancer group have 81.3%(13/16) of positive rate that is higher than in poorly differentiated group(37%(17/46)), which is statistically significant(P < 0.05), In addition, the positive rate of E- cadherin in gastric carcinoma in the depth of T1 + T2 group are 66.7%(16/24) which is higher than that in the T3+ T4 group(36.8%(14/38)). No lymph node metastasis in lymph node metastasis has a positive rate of 65.2%(15/23), higher than in the group with lymph node metastasis(38.5%(15/39)). In distant metastasis in no distant metastasis group has a positive rate of 59.6%(28/47), which is higher than in the distant metastasis group(13.3%(2/15)), in the TNM staging I+II period group has a positive rate of 65.6%(21/32), which is higher than III +IV group(30.0%(9/30)). All p values < 0.05 above. And E- cadherin has no relationship with gastric cancer patient's gender, age, tumor diameter or tumor site(P > 0.05).(3)Ki- 67 in normal gastric tissues, positive rate was 5%, below the gastric cancer group, with significant difference(P < 0.05). Ki- 67 expression in gastric cancer shows its in gastric carcinoma in the depth of T1 + T2 group are 41.7%(10/24) which is lower than that in the T3+T4 group(73.7%(28/38)). In lymph node metastasis in lymph node metastasis has a positive rate of 74.4%(29/39) higher than in the group without lymph node metastasis group(39.1%(9/23)), in the TNM staging III +IV period group has a positive rate of 80.0%(24/30), which is higher than I+II period group(43.8%(14/32)), All p values < 0.05. And Ki- 67 has no relationship with gastric cancer patient's gender, age, tumor location, tumor diameter, degree of differentiation, distant metastasis.(P > 0.05).(4)E-cadherin in gastric cancer tissue samples and the expression of Ki-67 after using SPSS22.0 Spearman rank correlation correlation analysis: E-cadherin and negatively correlated with the expression of Ki-67, including rs value is-0.291, and p values < 0.05.Conclusion:(1)E-cadherin expression in gastric cancer has a negative relationship with gastric cancer invasion depth, lymph node metastasis, distant metastasis, differentiation, TNM staging,reducing their expression in gastric cancer easily cause gastric cancer cells have aggressivity, more lymph node metastasis and distant metastasis.(2)Ki- 67 express suggest Ki- 67 in gastric cancer has a positive relationship with gastric cancer infiltration depth, lymph node metastasis and TNM stages, also shows the positive expression of Ki- 67 in gastric cancer is associated with the malignant degree and metastasis of gastric cancer.(3)E-cadherin in gastric cancer has a negative relationship with with the expression of KI-67.
