Objective:To research the TROP2 of the expression and clinical pathophysiological features in Non-small cell lung cancer Methods:1. By quantitative real-time PCR for detection the expression level of TROP2 mRNA in 20 NSCLC patients and adjacent normal tissues2. Utilizing enzyme linked immunosorbent assay(ELISA) kit to detect the serum concentration of TROP2 in 96 the patients with NSCLC or 20 benign lung disease Correlations of TROP2 with clinical pathological features were subsequently assessed3. TROP2 expression in 51 ADC tissues and 20 adjacent normal Lung tissues was detected by immunohistochemistry, Correlations of TROP2 with clinical pathological features were subsequently assessed Results:1. RT-PCR to detect NSCLC of TROP2 mRNA expression, the results displaied that the expression of TROP2 level in NSCLC was markedly higher than normal biliary tissues(P<0.05).TROP2 expression in squamous cell carcinoma is higher than adenocarcinoma of the lung(P<0.05).2. The level of TROP2 in serum of patients with NSCLC was dramatically higher than benign lung disease patients,the difference was dramatically significant(P <0.05). In NSCLC,serum of patients with TROP2 in SCC is higher than ADC,the differences were all significant(P<0.05). In serum of patients with NSCLC TROP2 levels and the degree of tumor differentiation,TNM stage differences with significant(P <0.05). N1-N3(lymph node metastasis)is higher than N0(No lymph node metastasis)patients TROP2 levels,although not statistically significant(P>0.05),but the serum of patients with lymph node metastasis of lung cancer TROP2 higher than without lymph node metastasis.There was no significant difference of cancer patient's age,gender and tumor size in serum of patients with NSCLC3. Immunohistochemistry analysis showed TROP2 next to ADC rare expre-ssion,positive expression was 10%(2/20);High TROP2 expression by immune-histochemistry was found in 28(54.9%) of the 51 tumor specimens,TROP2 of the expression was dramatically higher than adjacent normal tissues(P <0.05).There was significant correlations were found between TROP2 expression and histological differentiation, tumor TNM stage and lymph node metastasis(P=0.046?P=0.008?P=0.04)in ADC.we found that TROP2 expression patients and p atients age, gender and tumor size(P = 0.244,P = 0.964,P = 0.801) has no statistically difference; TROP2 tended to be expressed in cases with an unfavor able outcome. But no difference was found the correlation between TROP2 expression and months after surgery of Lung adenocarcinoma(P=0.35). Conclusions:1. TROP2 expression in NSCLC tissues was dramatically higher than in adjacent normal tissues,including squamous cell carcinoma was significantly higher than adenocarcinoma of the lung.2. In the serum of patients with NSCLC TROP2 levels were dramatically higher than in patients with benign lung lesion,the serum of patients with squamous cell lung TROP2 higher than in patients with adenocarcinoma of the lung; serum of patients with NSCLC TROP2 differentiation level of tumor,tumor staging difference was statistically significant,but not with paitents of age,gender,tumor size and lymph node metastasis had no dramatically correlation.3. TROP2 in lung adenocarcinoma tissue increased, there were conspicuously different compared with the expression of paracancerous tissue;The expression of TROP2 and tumor differentiation degree,lymph node metastasis and the TNM staging were positively correlated, but with patients of age, gender, tumor size, has no correlation;...
