| Objective: To investigate the effect of small interfering RNA(siRNA) targeting Trop2on the proliferation and invasion of lung adenocarcinoma H460cells.Methods: The recombinant adenovirus expression vector,which contained smallinterfering RNA (siRNA) targeting open reading frames of Trop2(rAd5-siTrop2),wastransfected into lung adenocarcinoma H460cells. Three groups were created for this study.Ctrl group, rAd5-siCtrl group(native control), rAd5-siTrop2group(knock down Trop2gene). Trop2mRNA and protein expressions were detected by Real-time PCR andWestern blotting analysis.The expressions of Cyclin Dl and P-ERK-1was also detected byWestern blotting analysis, respectively. The effect of Trop2on proliferation and invasionof lung adenocarcinoma H460cells were investigated using MTT and Transwell assay.Results: Trop2targeted small interfering RNA recombinant plasmids were constructedsuccessfully.Recombinant adenovirus vector rAd5-siTrop2significantly down-regulatedTrop2mRNA and protein expressions in lung adenocarcinoma H460cells;Cyclin D1andP-ERK-1was down-regulated simultaneously.Following silence of Trop2, cellproliferation and invasion of lung adenocarcinoma H460cells were reduced.Conclusion: SiRNA targeting Trop2can significantly down-regulate the expression ofTrop2in lung adenocarcinoma H460cells, and inhibit the proliferation and invasion oflung adenocarcinoma H460cells in vitro.... |
