| Background With an aging population issues highlighted,Alzheimer's disease(AD)has drawn increasing attention,because the disease is a progressive development of a serious impact on patients' memory and cognitive abilities.Western medicine treatment of multiple brain to protect the main,but often have a variety of side effects;Traditional Chinese medicine treatment mild or no side effects,in addition to improving intelligence can also improve other symptoms,so the combination therapy can play a role in each other,both improved intelligence can improve the efficacy of treatment and reduce adverse reactions.Based on this,the present study to investigate the electro-acupuncture combined with piracetam therapeutic effect of Alzheimer's disease in order to provide experimental data for clinical application.Objective To observe the effect of acupuncture combined with piracetam induced by amyloid beta(Abeta 25-35)SD model of Alzheimer's disease in rats with memory,cognitive ability and 5-HT and NE levels of monoamine transmitters were influenced by the study,to investigate the effect of electro acupuncture combined with piracetam of Alzheimer disease(AD),the therapeutic effect,for the future of ad of traditional Chinese medicine and Western medicine combined with treatment to explore new methods and directions.Method 50 SD rats using a random number table method into blank group 10,making a model group(n = 40)and the beta amyloid peptide fragment 25-35(beta amyloid peptide 25-35)bilateral hippocampal stereotaxic injection method of making rat model of Alzheimer's disease.The successful model of 40 SD rats were randomly divided into model group,electro acupuncture group,Derby lacidipine Tan group and EA combined with piracetam group according to.After third days,seventh days and twenty-eighth days after modeling,the learning and memory performance of rats in each group were tested by Y electric maze test.Made successful model from the second day of electroacupuncture combined with piracetam group every day to crawl fixed after each irrigation stomach piracetam suspension 1ml/D,and then impose "Baihui" and "Shenshu" "Zusanli" acupuncture gas connected with pulse electrotherapy instrument in a continuous wave,frequency of 150 Hz.In SD rat limbs occurred mild jitter for the proper stimulating intensity and exciting time every time 3~5 minutes,once daily,6 days for a course of treatment,a course after the end of the rest one day into the next course,a total of four courses of treatment.Piracetam group every day to crawl after fixation of each administered piracetam suspension 1ml/d,not the implementation of electro acupuncture therapy.Electric acupuncture group for 4 courses of treatment of simple electroacupuncture.The blank group and model group the same degree of grasping,fixed,neither gavage mixed piracetam suspension nor electroacupuncture therapy.4 courses of treatment after the end of the computer monitor and video recorder at the same time continuous monitoring and recording 12 hours(late 8:00 early 20:00-)within each group of SD rats the number of times and the time of awakening.After the completion of the above indexes were immediately in were isolated SD rat frontal lobe of the brain tissue,accurate weighing with tin foil,rapid cryopreservation to be measured,waiting for all the specimens were prepared after homogenization,centrifugation,the supernatant was obtained and detected in 50 SD rat frontal 5-HT of NE,Da,frontal dopamine monoamine neurotransmitter levels.Result 1.effect on the memory capacity of SD rat: by Y-type maze test observation of 5 groups of SD rats with cognitive behavior and the ability of learning and memory and understanding to modeling the 3D,five groups of SD rats learning memory ability had no significant difference.7 days,model group,electro acupuncture group,Derby lacidipine Tan group and EA combined with piracetam group four groups of SD rats showed learn to escape the required electrical stimulation times decreased,and the number of error reduction,decrease the abilities of learning and memory,the 4 groups after modeling SD rats respectively,compared to the blank group,the differences were statistically significant(P < 0.05);also indicated that rats in the model.28 d,no statistical significance of EA group and piracetam group compared(P>0.05).Electro acupuncture combined with piracetam group were differences were statistically significant(P < 0.05)compared with piracetam group,electroacupuncture group,electroacupuncture combined with piracetam group SD rats learning and memory ability has improved.Effects of electroacupuncture combined with piracetam group,electroacupuncture group,piracetam group respectively with model group compared to the differences were statistically significant(P < 0.05),electroacupuncture combined with piracetam group,electroacupuncture group,piracetam group SD rats were learning memory ability were improved.Electro acupuncture combined with piracetam group and blank group,difference has statistical significance(P < 0.05),electroacupuncture combined with piracetam group has not yet reached the complete recovery of the treatment effect.2.the influence of the awakening ability of SD rats: after the end of the 4 treatment period,the video recorder was used for 12 hours(20:00-8:00)for hours after the end of treatment,and the sleep state of SD rats was continuously observed.And blank group,compared with the model group,electro acupuncture group,Derby lacidipine Tan group and EA combined with piracetam group 4 groups of SD rats at night the activity times reduction and awakening time is shortened and the 4 groups after modeling SD rats respectively,compared to the blank group,the differences were statistically significant(P < 0.05);also shows once again that the successful modeling.No statistical significance in the EA group and piracetam group compared(P>0.05).Electro acupuncture combined with piracetam group were differences were statistically significant(P < 0.05)compared with piracetam group,electroacupuncture group electroacupuncture combined with piracetam group SD rats nighttime activities significantly increased the number,awakening time was significantly prolonged.Effects of electroacupuncture combined with piracetam group,electroacupuncture group,piracetam group respectively with model group compared to the differences were statistically significant(P < 0.05),electroacupuncture combined with piracetam group,electroacupuncture group,piracetam group SD rats were nocturnal activity frequency increased significantly,awakening time was significantly prolonged.Electro acupuncture combined with piracetam group and blank group,difference has statistical significance(P < 0.05),electroacupuncture combined with piracetam group has not yet reached the complete recovery of the treatment effect.3.the levels of monoamine transmitters in frontal lobe of SD rats in each group.Compared with the control group,the intervention treatment model group,electro acupuncture group,Derby lacidipine Tan group and EA combined with piracetam group SD rat frontal cortex tissue in the level of 5-HT,DA and NE levels were significantly reduced,the 4 groups after modeling SD rats respectively,compared to the blank group,the differences were statistically significant(P < 0.05)and EA group and piracetam group had no significant difference(P > 0.05).Electro acupuncture combined with piracetam group were differences were statistically significant(P < 0.05)compared with piracetam group,electroacupuncture group,electroacupuncture combined with piracetam group SD rats level of 5-HT,DA and NE levels were significantly increased.Effect of electroacupuncture combined with piracetam group compared with control group the difference was statistically significant(P < 0.05).Effects of electroacupuncture combined with piracetam group,electroacupuncture group,piracetam group respectively with model group compared to the differences were statistically significant(P < 0.05),electroacupuncture combined with piracetam group,electroacupuncture group,piracetam group SD rats level of 5-HT,DA and NE levels were significantly increased.Effect of electroacupuncture combined with piracetam group compared with control group the difference was statistically significant(P < 0.05).Conclusion electroacupuncture at Baihui,Shenshu,Zusanli combined with piracetam can significantly improve memory of SD rats,cognitive ability and frontal lobe of brain 5-HT and NE levels of monoamine transmitters,SD rat spatial memory ability,improve rat arousal state,improve the frontal lobe monoamine neurotransmitter levels. |
