Objective The antiviral effect of shen qi zhi pao granule is investigated by establishing the animal models of guinea pigs infected with HSV-2 in this paper.Serum levels of cytokines,protein and gene expression by normal healthy guinea pigs,HSV-2 infection model and the treatment group will be detected and compared,and then we can make a analysis of the role of cytokines in the pathogenesis of genital herpes.The analysis of viral titer,histopathology and observation of ultrastructure in the vulva skin epithelial cells and ganglion will be carried out at the same time.All the works is to providing a reliable objective basis for clinical practice of Chinese medicine and supply scientific experimental basis for themodern medicine exploitation and the utilization of traditional Chinese medicine at the same time.Methods This experiment generate a animal model by Hartly Dunkin white female guinea pigs which genital tract infected with HSV-2 and were randomly divided into four groups :normal control group,model group and Shenqi zhipao granule group and acyclovir(ACV)group.Each group was given corresponding drug therapy(model group and normal control group were given normal saline).The skin symptoms of guinea pig are observed and compared with the microscope and determination of viral titer in the vaginal tissue of guinea pigs by cell culture as well as HE staining for the production of the uterus? fallopian tubes? ovarian tissue pathological sections in the study.We observe and record the pathological results and the changes of ultrastructure in vulva skin epithelial cells and spinal ganglion.Geneexpression of IL-2?IL-6?IFN-??TNF-??NF-?B mRNA in the uterus and ovary of each group were determined by RT-PCR.There is an important step that detecting the content of IL-2?IL-6?IFN-??TNF-? in the serum by ELISA,using western blot to detect the expression of NF-?B in guinea pig uterus should be included.All the data above were statistically analyzed.Results At the end of the treatment,check the virus titer inside the vagina of guinea pig and record symptom scores of the vulva skin so as the tissue pathology of uterus,tubal,ovarian and ultrastructure from vulva skin epithelial cells and spinal ganglion.There is no significant difference between Shenqi granule group and aciclovir group(P>0.05)but a significant difference between the treatment group and model group(P<0.05).Compared with the model group,mRNA expression of IL-2?IFN-? in the uterus and ovary tissue of the guinea pig by shen qi zhi pao granule group results in a significant up-regulation,IL-6 ? TNF-? and NF-?B mRNA in down regulation,The difference is statistically significant(P<0.05).High IL-2 and IFN-? content in serum but IL-6 and TNF-? content is low,the difference was statistically significant(P<0.05).Low expression of NF-kB protein in guinea pig uterus,the difference is statistically significant(P<0.05).There is no significant difference between the Shenqi zhipao granule group and acyclovir group(P>0.05).Conclusion We find that shen qi zhi pao granule has a strong effect against HSV-2 whose therapeutiu effect is equal to Acyclovir.Shen qi zhi pao granule reduces the inflammatory injury of uterine and ovarian tissues of the guinea pig infected with HSV-2 by regulating the mRNA expression of IL-2?IL-6?IFN-??TNF-??NF-?B so as to protect the body and achieve the antiviral effect finally.
