Objective:To research the tomato leaves(the perennial herbaceous tomato that means Ly-copersicon esculentum Mill leaves).The eluting power of AB-8 macroporous resin and D-101 macroporous resin were investigated to contrast the adsorption ability of n-butyl alcohol extract and analysis their separating effect of antioxidant ingredient.The tomato leaves of n-butyl alcohol part was extracted and done systematic experiment of the chemical composition to reveal the chemical material foundation of antioxidant effect.for the sake of selecting appropriate macroporous resin,providing a scientific basis for its further study,development,utilization and furnishing industrial production and application with theoretical reference.Methods:Tomato leaves were percolated and extracted to isolate parts of n-butanol,then the AB-8 macroporous resin and the D-101 macroporous resin were used to elute the n-butyl alcohol extract part of tomato leafs.Reducing ability,Clearance rate of oxygen radicals and hydroxyl radical were examined by the methods of Prussian bilue method,pyrogallol autoxidation method and phenanthroline-Fe2+oxidation method to compare the antioxidant capacity of different elution gradient.Ultimately compare the adsorption separation effect of thus two macroporous resin.The effective compound was isolated and purified from n-butyl alcohol extract part of tomato leafs by the silica gel column chromatography,polyamide column chromatography,solvent extraction,recrystallization repeatedly and so on.Physicochemical properties and spectral analysis method such as ESI-MS,EI-MS,ESI,1H-NMR,13C-NMR were utilized to identify the structures of the compounds.Results:Different gradient elution of n-butyl alcohol extract part of tomato leafs eluted separated by AB-8 and D-101 macroporous resin are all have obvious anti-oxidation,and the anti-oxidation are growing with the concentrations increasing;Among them,antioxidant effect of the 30%gradient and 50% gradient of n-butyl alcohol extract part of tomato leafs eluted separated by AB-8 and D-101 macroporous resin were more strong than 80% gradient and water gradient.Eleven compounds were isolated from the n-butyl alcohol extract part and ten of them were identified,they are tomatine(1),tomatidine-3-O-?-D-glucopyranoside(2),soladulcidine-3-O-?-Dglucopyranosyl-(1?3)-?-D-glucopyranosyl(1?4)-?-D-galactopyranoside(3),Solalyratine B(4),soladulcidine-3-O-?-lycotetraoside(5),quercetin(6),?-sitos terol(7),?-spinasterol(8),n-butyl-O-?-D-fructopyranoside(10)and rutin(11).Conclusion:Both AB-8 macroporous resin and D-101 macroporous resin can be used for the separation and purification of the n-butyl alcohol extract part of tomato leafs.The 30% gradient and 50% gradient of n-butyl alcohol extract part of tomato leafs eluted separated by AB-8 and D-101 macroporous resin have more antioxidant ingredient than than 80% gradient and water gradient;Compounds of 2,3,4,5 and 8 were firstly isolated from this plant.
