Objective:Base on the orthogonal test,to investigate the the best compatibility rules and hypoglycemic effect of Ye's Xiaoke recipe.Methods:1.Rat models of type 2 diabetes were made by intraperitoneal injecting low-dose steptozotocin accompanied by feeding high-lipid and high-sugar diet.Based on orthogonal design,mice was divided into 18 groups,12 in each group according to L18(37)table.Then the rats were given Yeshi xiaokefang 14 day,and fasting blood glucose(FBG),Glucagon(GC),insulin(Ins),triglyceride(TG),total cholesterol(TC),high-density lipoprotein cholesterol(HDL-c)and low-density lipoprotein cholesterol(LDL-c)were as indicators.The synthesis score of relevant multiple indicators was calculated and the multiple indicators were analyzed by using orthogonal design and variance analysis.2.Verification experiments in the same way replication model of diabetic rats,were divided into 6 groups,namely the best compatibility of high,low dose group,model group,metformin group and blank group,serum fasting blood glucose(FBG),serum insulin(INS)and glucagon prime(GC)and Gan oil triglyceride(TG),total cholesterol(TC),high density lipoprotein(HDL-C),low density lipoprotein(LDL)levels of the indicators,also under the microscope observation pathological changes of pancreatic tissue,further verified and analyzed Ye Xiaoke recipe and the best prescription of anti diabetic action.Results:1.In orthogonal test,compared with the model group,the test group,in varying degrees to reduce animal blood glucose(FBG),glucagon prime(GC),triglyceride(TG),total cholesterol(TC),low density lipoprotein(LDL),serum insulin(INS)and high density lipoprotein(HDL-C)also has certain promotion;The range analysis of the comprehensive evaluation of the indicators showed that primary and secondary subsequence were C>B>A>D>E>F;analysis of variance showed that gypsum,Anemarrhena rhizome,rehmannia root,ophiopogon root,Radix Paeoniae Alba and licorice the six factors on the comprehensive index of the type 2 diabetes have varying degrees of impact,but the extent of influence have great difference,the optimum of Yeshi xiaokefang was as follows:30 g Radix Rehmanniae;9 g Armeniacae Common Anemarrhea;and 30 g Gypsum Fibrosum;2 g Glycyrrhiza;9 g white peony root;and 6 g tuber of dwarf lilyturf.2.In the verification experiments,each dose group and model group of Yip's diabetes the best compatibility,can significantly reduce the level of model rats' blood glucose(FBG),total cholesterol(TC),low density lipoprotein(LDL);high dose and middle dose group of serum insulin(INS)levels increased significantly,the carotene(GC)in dose group of glucagon decreased;high dose and middle dose groups of glycerin three greases(TG)increased significantly,also in each dose group can impaired diabetic rat pancreatic tissue structure was improved to different degrees.Conclusion:1.The effect of Yeshi xiaokefang on hypoglycemic is optimal in the dosage of 30g Gypsum Fibrosum;9g Armeniacae Common Anemarrhena;and 30 g Radix Rehmanniae;6 g tuber of dwarf lilyturf;9 g white peony root;and 2 g Glycyrrhiza.2.Its hypoglycemic mechanism of which probably lies in the modification of carbohydrate metabolism,accelerating the repair of pancreatic inlet ? cells,increasing secretion and decreasing decomposition of insulin,regulating lipids metabolism to improve insulin resistance.
