PD-1 Expression On PBMCS,CD3+T Cells And CD8+T Cells Impact Phosphorylation Of P38 MAPK,NF-?B During HIV-1 Infection

Objective In this study,we investigated the levels of PD-1,NF-?B,p38 MAPK on PBMCs,CD3+T cells and CD8+T cells during HIV-1 infection,and the correlation of PD-1 and the phosphorylation levels of NF-?B?p38 MAPK.Our goal is to explore what is the influence of PD-1 on PBMCs,CD3+T cells and CD8 +T cells in HIV-1 infected patients.Method 39 patients with HIV-1 infection were enrolled in our study and divided into two groups including 15 Typical AIDS patients and 24 HIV-1 Carriers.And 9 healthy donors were recruited in our study.All the individuals who participated in this study provided written informed consent.PBMCs isolated by gradient centrifugation with Ficoll-Hypaque,after washing with RPMI-1640,were divided into A,B and C tube,single-cell suspension of 1×106 cells per tube.A tube was stained with CD3,CD8 and PD-1.B tube and C tube were stimulated by Purified anti-human CD3 and Purified anti-human CD28.Then,the cells were fixed.B tube was stained with anti-human CD3,anti-human CD8,anti-human PD-1 and anti-human p38 MAPK.C tube was stained with anti-human CD3,anti-human CD8,anti-human PD-1 and anti-human NF-?B.The frequencies of PD-1,p38 MAPK and NF-?B on PBMCs,CD3+T cells and CD8+T cells were assessed by flow cytometry.We further analyze the relationship about the expression of,p38 MAPK and NF-?B on PBMCs,CD3+T cells and CD8+T cells during HIV-1 infection.Results:1.Compared with TPs,the frequencies of PD-1 on PBMCs were significantly lower in HDs and CRs(P<0.05,P<0.05).Compared with CRs,the frequencies of p38 MAPK on PBMCs were significantly higher in TPs and HDs(P<0.05,P<0.05).Compared with TPs,the frequencies of NF-?B on PBMCs were significantly higher in HDs and CRs(P<0.05,P <0.05).Correlation analysis showed that there is negative correlation between CD4 count and p38 MAPK expression on PBMCs in HIV-1 infection(P<0.05),and there is negative correlation between CD4 count and PD-1 expression on PBMCs in HIV-1 infection(P<0.05),and there is positive correlation between CD4 count and NF-?B expression on PBMCs in HIV-1 infection(P<0.05).There is positive correlation between p38 MAPK expression and PD-1 expression on PBMCs in HIV-1 infection(P<0.05),and There is negative correlation between NF-?B expression and PD-1 expression on PBMCs in HIV-1 infection(P<0.05).2.Compared with TPs,the frequencies of PD-1 on CD3+T cells were significantly lower in CRs and HDs(P<0.05,P <0.05).Compared with CRs,the frequencies of p38 MAPK on CD3+T cells were significantly higher in TPs and HDs(P<0.05,P<0.05).Compared with HDs,the frequencies of NF-?B on CD3+T cells in TPs and CRs were no different(P>0.05,P>0.05,P>0.05).Correlation analysis showed that there is negative correlation between CD4 count and p38 MAPK expression on CD3+T cells in HIV-1 infection(P<0.05),and there is negative correlation between CD4 count and PD-1 expression on CD3+T cells in HIV-1 infection(P<0.05),and there is positive correlation between CD4 count and CD3 expression on PBMCs in HIV-1 infection(P<0.05),and there is no correlation between CD4 count and NF-?B expression on CD3+T cells in HIV-1 infection(P>0.05).There is positive correlation between p38 MAPK expression and PD-1 expression on CD3+T cells in HIV-1 infection(P<0.05),and There is no correlation between NF-?B expression and PD-1 expression on CD3+T cells in HIV-1 infection(P<0.05).3.Compared with TPs,the frequencies of CD8 on PBMCs were significantly lower in CRs and HDs(P<0.05,P<0.05),compared with CRs,the frequencies of CD8 on PBMCs were significantly lower in HDs(P<0.05).Compared with TPs,the frequencies of PD-1 on CD8+T cells were significantly lower in CRs and HDs(P<0.05,P<0.05).Compared with CRs,the frequencies of p38 MAPK on CD8+T cells were significantly higher in TPs(P<0.05).Compared with HDs,the frequencies of NF-?B on CD8+T cells in TPs and CRs were lower(P>0.05,P>0.05,P>0.05),compared with CRs,the frequencies of NF-?B on CD8+T cells in TPs was lower(P>0.05).Correlation analysis showed that there is negative correlation between CD4 count and p38 MAPK expression on CD8+T cells in HIV-1 infection(P<0.05),and there is negative correlation between CD4 count and PD-1 expression on CD8+T cells in HIV-1 infection(P<0.05),and there is negative correlation between CD4 count and CD8 expression on PBMCs in HIV-1 infection(P<0.05),and there is positive correlation between CD4 count and NF-?B expression on CD8+T cells in HIV-1 infection(P<0.05).There is positive correlation between p38 MAPK expression and PD-1 expression on CD8+T cells in HIV-1 infection(P<0.05),and There is no correlation between NF-?B expression and PD-1 expression on CD8+T cells in HIV-1 infection(P<0.05).Conclusion 1.PD-1 is involved in immune response via positively regulated phosphorylation of p38 MAPK in PBMCs,CD3+T cells and CD8+T cell during chronic HIV-1 infection.2.PD-1 is involved in immune response via positively regulated phosphorylation of p38 MAPK in PBMCs,CD3+T cells and CD8+T cell during chronic HIV-1 infection.3.PD-1 is involved in immune response via negatively regulated phosphorylation of NF-?B in PBMCs and CD8 + T cell during chronic HIV-1 infection.
