Objective: To investigate the effect of Benner Pury on electrocardiogram in diabetic rats and the intervention effect of the remodeling of the left ventricular gap junction protein Cx43.Methods: 1.48 male SD rats were randomly divided into control group(N=12)and diabetic group(N=18 group)and Benner Pury(N=18).After one week adaptive feeding,diabetic group and Benner Pury group were given streptozotocin(STZ,65mg/Kg)to build type I diabetes model through disposable left lower abdominal injection,,Benner Pury were given the 10mg/kg/d gavage intervention,diabetes control group given saline with the same dose every day to intervene,the rats were killed after 8 weeks,observe the general condition of rats(blood glucose,body weight,heart index),electrocardiogram(ECG arrhythmia,the main parameters including heart rate,PR interval,QTC QTD,etc.),myocardial morphology,distribution and expression of connexin Cx43 in the left ventricle.During the experiment,the rats were excluded from the model failure,death,decreased blood glucose and so on,and finally included in the statistical analysis: blank control group(n=12),diabetes control group(n=12),Benner Pury group(n=11).2.the main parameters of electrocardiogram(including heart rate,PR interval,QTC,QTD)and arrhythmia were recorded by using physiological recording instrument.3.The morphological changes of myocardium were showed by HE staining.4.The spatial distribution of Cx43 in each group was revealed by immunofluorescence technique.5.to detect the expression of gap junction protein P-Cx43 and Cx43 in each group by Western Blotting6.Data processing: research data were expressed to X ± s by SPSS software for statistical analysis,measurement data by is normality test and homogeneity of variance test using analysis of variance,group pairwise comparison with Q test,with alpha =0.05 as the standard test,P < 0.05 for differences have statistical significance.Results: 1.Diabetic model group appeared typical "symptoms,some rats appeared diabetes related complications such as cataract,even death.Diabetic group compared with blank control group rise in blood sugar,weight loss,cardiac index increased(P <0.05)and benazepril group compared with the diabetic control group cardiac index had significant difference(P < 0.05)2.In the diabetic control group various forms of arrhythmia was observed,diabetes group compared with blank control group decreased heart rate,prolong the QTc and QTd increased(P < 0.05)and benazepril group and the diabetic control group in heart rate,QTc,QTd also show statistically significant(P < 0.05)3.HE staining: blank control group,myocardial arrangement rule,myocardial mass without obvious hyperplasia.Diabetic control group myocardial fibers arranged disorderly,fuzzy boundaries of nucleus and cytoplasm.Benazepril group compared with diabetic control group myocardial arrangement,myocardial interstitial hyperplasia improved.4 munohistochemical staining showed: blank control group,myocardial Cx43 distribution in the intercalated discs,diabetic control group,Cx43 distribution in the side of the myocardial cell membrane.Benazepril group Cx43 distribution is complex,distributed in intercalated discs,aslo distributed in the cell membrane of side.5.Western Blotting showed: compared with the blank control group,diabetic control group left ventricular myocardial tissue Cx43,P-Cx43 expression was reduced,the difference was statistically significant(P < 0.05).Conclusion: 1.serious damages produced by Diabetes on rat myocardium,myocardial structure: myocardial degeneration,necrosis,between qualitative fibrosis(HE staining).Myocardial function: the emergence of the various forms of arrhythmia,heart rate decreased,repolarization time delay,QT dispersion increase(ECG),diabetes on rat ECG has a bad influence,the occurrence of diabetes may be associated with arrhythmia has a close relationship.2.benazepril can improve diabetic rats myocardial degeneration,necrosis,fibrosis(HE staining),improve the heart rates of diabetic rats decreased,prolongs repolarization,QT dispersion increase may plays an important protective role in diabetes induced arrhythmia.3.Normal rat left ventricular gap junction Cx43 protein distribution in the intercalated discs,diabetic rats left ventricular exists Cx43 protein reconstruction(Cx43 decreased expression and structure exist side distribution)to participate in the occurrence of ventricular arrhythmias in diabetic rats4.Benner Pury may have an anti-ventricular-arrhythmia effect,and its partial mechanism may be achieved through improving the reconstruction of gap junction protein Cx43,which still needs further proof.
