Objective:Examine the protective effect of Benazepril on cardiac function in diabetic rats.METHOD:Induce diabetic model in Sprague-Dawley(SD) male rats by intraperitoneal injection of streptozotocin(STZ). The successful model of diabetic rats were randomly divided into diabetic group and benazepril group, the control group were rats that were not injected with STZ. The control group and diabetic group were fed with tap water by gavage and the benazepril group with benazepril powder dissolved tap water. After 8 weeks.cardiac ultrasound was used to measure left ventricular end diastolic diameter (LVIDD). left ventricular end systolic diameter (LVIDS). ejection fraction (EF). left ventricular fraction shortening (FS). And left ventricular peak pressure (LVSP). left ventricular end diastolic pressure (LVEDP). maximal rates of pressure development and decline were measured by ventricular intubation. And heart weight index was also measured. RESULT:Echocardiography shows:In the diabetic group compared with the control group, LVIDD [(6.79±0.58) vs (7.73±1.19) mm, p<0.05], heart rate [(350±54) vs (432±44) bpm, p<0.01], FS [(37.15±8.81) vs (52.04±9.68)%, p<0.01], EF [(0.72±0.11) vs (0.86±0.09), p<0.01] were significantly decreased (p<0.01); Compared with the diabetes Group, LVIDS of benazepril group was significantly decreased [(3.74±0.64) vs (4.25±0.53) mm, p<0.05], heart rate [(382±23) vs (350±54) bpm, p<0.05], EF [(0.80±0.08) vs (0.72±0.11), p<0.05], FS [(0.80±0.08) vs (37.15±8.81)%, p<0.05] were significantly increased. Hemodynamics shows:In the diabetic group compared with the control group,+dp/dt [(4044.66±1318.27) vs (6015.63±905.70) mmHg/s, p<0.05] was decreased significantly. And LVSP, LVEDP,-dp/dt show no significant differences; Compared with diabetes group,+dp/dt of benazepril group increased significantly [(5424.27±923.68) vs (4044.66±1318.27) mmHg/s, p<0.05]. But LVSP, LVEDP,-dp/dt shows no significant differences. Compared with the control group, body weight of diabetic group decreased significantly [(221±46) vs (421±62) g. p<0.01], with the heart weight index significantly increased [(4.07±0.67) vs (3.23±0.14) mg/g, p<0.05]; Compared with diabetic group, body weight and heart weight index of benazepril group shows no significant differences.CONCLUSIONS:Cardiac insufficiency was found in STZ-induced diabetic rats at eighth week, benazepril shows a protective effect on the cardiac function of diabetic rats.
