Objective: Intracranial tumor is one of the most common diseases in neurosurgery disease,including glioma,meningioma,pituitary adenoma,acoustic neuroma,brain metastases,epidermoid cyst,etc.,in which the incidence of gliomas is the highest.The incidence of glioma not only remains high but its oncocyte also has biodiversity,strong invasion and infiltration ability and also transitivity,so Glioma is still the most commonest central nervous system tumor with the worst prognosis after comprehensive treatment.In recent twenty years,even comprehensive treatment like operation treatment and chemotherapy can still not improve the median survival time of patients evidently or delay the recurrence.Therefore,preopreative examination is of great importance to predicate the pathology level of Glioma and determine the Glioma boundary and other information about Glioma.IL-12 is mainly secreted by antigen presenting cells such as mononuclear macrophage,B cells and dendritic cells(DC)and it has many kinds of bioactive cytokines.It plays an irreplaceable role in the antitumor immunity.In this paper,ELISA method was adopted to measure the IL-12 content in the serum of 8 normal individuals and the serum of 23 patients with glimoa before operation.Researches were made on the relation among VEGF expression levels in the tumor specimens of glioma patients that were obtained by immunohistochemical method and glioma high-low levels and IL-12.It is hoped that an easy,accurate and specific method can be found to predicate the high-low level of glioma before operation,so as to assist in surgery of glioma and postoperative chemoradiation therapy.Methods:Twenty-three patientss with primary glioma that had received operation treatment in the neurosurgery department of Second Hospital of HebeiMedical University from September 2015 to December 2015 were randomly included in this experiment.They were classified according to WHO 2007 standards and WHO I,II levels were divided into the low level glioma group,totally 12 cases.WHO III-Ⅳ levels were divided into high level glioma group,a total of 11 cases.Before operation,5ml blood specimen was taken from each patient under the consent of patients and their family members and the specimens were stored in-80 ℃ refrigerator after standing and centrifugation.During the operation,glioma specimen was taken at the site near to tumor center(the tumor center should be confirmed by experienced pathology teacher,neurosurgery teacher and the operator),about 1×1×1cm,and then it was fixed and sealed by formalin solution of 100 mmol / L,waiting to be made into wax samples.Eight healthy individuals were randomly selected from people that had received physical examination in the physical examination department of our hospital and blood drawing,standing,centrifugation and storage were conducted as the same with the glioma group.ELISA method was used to measure IL-12 concentration and Immunohistochemical method was used to detect VEGF expression level in the tumor sample.SPSS 19.0 software was adopted for data analysis.The measurement data that were in accordance with normal distribution were presented in mean ±standard deviation and the non-normal data was presented in median(quartile).The difference between the normal group and the glioma group in serum IL-12 level were compared by t test.Wilcoxon rank sum test was used to compare the glioma low level group and high level group.Wilcoxon rank sum test was used to compare VEGF expression between low level group and high level group.Rank correlation was used to discuss the relation between IL-12 level and VEGF expression.Results:1.Comparison in serum IL-12 level between the normal control group and the glioma group: the first part of the experiment determined the serum IL-12 level of 8 healthy individuals in the control group and 23 gliomapatients.Testing results: the content of IL-12 in the serum of healthy adults was 19.680 ± 10.138pg/ml and that in the serum of glioma patients was27.858 ± 2.714pg/ml.Both of them accorded with the normal distribution and the variance was not equal.T test was used here(a=0.05),testing results:t′=-2.254.The two-two comparison difference P were all higher than 0.05 and there was no statistical difference.The content of IL-12 in the glioma low level group was 30.091(2.038)and that in the glioma high level group was25.814 ± 1.374.Wilcoxon rank test was used here(a=0.05),testing results:Z=-3.200.The two-two comparison difference P were all lower than 0.05 and there was statistical differnce.2.The relation of VEGF expression level in the tumor tissue of glioma patients and the high-low level of glioma: the second part of the experiment determined VEGF expression level in the tumor tissue of 23 glioma patients.The expression percentage of VEGF in the low-level glioma tissue was 24.700(3.71),the expression percentage in the glioma tissue of high-level group was40.670(21.10).Wilcoxon rank sum test was adopted in this test,results:Z=-4.062,P=0.001 and it had statistical significance.3.The relation of serum IL-12 concentration and VEGF expression level in the glioma tissues in patients with glioma: rank correlation analysis was conducted on serum IL-12 concentration and VEGF expression level in the glioma tissues.Since the data related to IL-12 concentration accorded with the normal distribution while the data of VEGF expression level did not accord with the normal distribution,therefore,rank correlation analysis rs=-0.753,P=0.001<0.05 and they had significant negative correlation.Conclusions:Conclusions: there is no significant difference in serum IL-12 concentration between the healthy individuals and the glioma patients and there was no statistical significance,but there was difference in the serum IL-12 concentration between low grade glioma group and high grade glioma group and it had statistical significance.Therefore,IL-12 concentration in the serum of patients cannot be used to diagnose Glioma in the early stage,but itcan be the index to predicate the high and the low grade Glioma before operation.
