Objective: Use of immunohistochemistry(IHC)and Western blot(WB)to research the exoression of RAF,MEK and ERK in the two group of invasive pituitary adenoma(IPA)/ non-invasive pituitary adenoma(NIPA)and functional pituitary adenoma(FPA)/ no-functional pituitary adenoma(NFPA),analysis the difference of RAF,MEK and ERK between the two kinds of group above and clarify the role of this channel in pituitary adenoma(PA)further,in order to find a new target for the treatment of PA.Methods:60 cases of sample which were resected by nasal operation at the second hospital of Hebei Medical University from June 2014 to December 2015 were selected.These samples were grouped into the invasive pituitary adenoma / non-invasive pituitary gland and functional pituitary adenoma / pituitary adenoma(see Table 1).The expression of RAF,MEK and ERK in the group of invasive pituitary adenoma / non-invasive pituitary gland and functional pituitary adenoma / pituitary adenoma were detected by Western blotting and immunohistochemistry.Immunohistochemical light microscope was analysisde by two pathologists using double blind method reading and scoring double semi quantitative method score.The RAF,MEK and ERK positive expression rate were statisticsed by the score of the statistics;Western blot analysis: the RAF,MEK and ERK relative values were calculated by compared the RAF,MEK and ERK gray value and the?-actin gray value which were analysised by the labwork analysis software.The count data was by chi square test,and the measurement data which be expressed by mean ± standard deviation(Sx±),wes statistical analysised by two independent sample t test.All statistics were analysised by IBM SPSS Statistics 19 software.If P<0.05 the differences were considered statistically significant.Result: 1 the result of immunohistochemistry 1.1 The comparison results between invasive group and non-invasive group resultsThe positive expression and positive expression rates of RAF,MEK and ERK in the 30 cases of invasive group were 28(93.33%)cases,26 cases(86.66%)and 27 cases(90%).The positive expression and positive expression rates of RAF,MEK and ERK in the 30 cases of non invasive group were 20 cases(66.66%),16 cases(53.33%)and 18(60.00%).The number of cases and the effective rate of invasive group were significantly higher than that of non-invasive group,and statistical analysis showed a significant difference between the two groups(P<0.05)(see Table 2).1.2 The comparison results between functional group and non-functional group resultsThe positive expression and positive expression rates of RAF,MEK and ERK in the 28 cases of functional group were 22(78.57%),19(67.85%)and 23(82.14%);The positive expression and positive expression rates of RAF,MEK and ERK in the 32 cases of non-functional group were 26(81.25%),23(71.87%)and 22(68.75%),respectively.The number of cases and the effective rate of invasive group were significantly higher than that of non-invasive group,and statistical analysis showed a significant difference between the two groups(P<0.05)(see Table 3).2 Western blot results 2.1 The comparison results between invasive group and non invasive group resultsThe three factor protein expression of RAF,MEK and ERK in the 30 cases of invasive group were0.7276±0.14067,0.2585±0.03339 and 0.3873±0.06493;The three factor protein expression of RAF,MEK and ERK in the 30 cases of non-invasive group were 0.1465±0.06036,0.1073±0.03601 and 0.2587±0.06493.The three factors were showed a significant difference between the two groups by statistical analysis.(P<0.05)(see Table 4).2.2 The comparison results between functional group and non-functional groupThe three factor protein expression of RAF,MEK and ERK in the 28 cases of invasive group were0.4316±0.30472,0.1837±0.08288 and 0.3226±0.105715;The three factor protein expression of RAF,MEK and ERK in the 30 cases of non-invasive group were0.4416±0.32394,0.1819±0.08581 and 0.3234±0.07986.There was no statistical significance between the two groups of the three factors.(P>0.05)(see Table 5).Conclusion:1 The expression of Raf,MEK and ERK in invasive pituitary adenoma were significantly higher than that in non-invasive pituitary adenoma.2 The expression of Raf,MEK and ERK were no significant difference between the two group of functional pituitary adenoma and non-functioning pituitary adenomas.3 Raf,MEK and ERK play an important role in the development and invasion of pituitary adenoma.4 The high expression of Raf,MEK and ERK in invasive pituitary adenoma provide a new direction evidence for treatment.
