Objective: To discuss the expression of tumor M2-pyruvate kinase(TuM2-PK),6-phosphofructo-2-kinase/fructose-2,6-bisphosphatase(PFKFB3)and transketolase-like gene 1(TKTL1)in laryngeal carcinoma and adjacent mucosa and its relationship with Clinical pathology.The mechanism and the interaction of the three factors during the occurrence,development and metastasis of laryngeal carcinoma were also explored.Methods: 36 cases of laryngeal carcinoma tissue and 20 cases of adjacent mucosa were chosen from the Second Hospital of Hebei Medical University from 2014/6 to 2015/12.All tumors tissues were pathologically confirmed as laryngeal squamous cell carcinoma,the patients had received neither chemotherapy nor radiotherapy before surgery.The expression of TuM2-PK?PFKFB3 and TKTL1 protein in 36 cases of laryngeal carcinoma tissue and 20 cases of adjacent mucosa were detected by immunohistochemical methord.The comparison of positive expression rate of protein between laryngeal carcinoma and adjacent mucosa was analyzed by Chi-square test.The relativity between the positive expression rate of protein and other clinicopathological parameters was analyzed by Fisher's exact probabilistic metho.The relationship among these three factors were analyzed by Spearman Rank Correlation Analysis.All data were analyzed by SPSS13.0 statistical software,with P<0.05 for a statistical difference.Results:1 The positive expression rate of TuM2-PK in laryngeal carcinoma tissue and adjacent mucosa was 61.11% and 25.00% respectively,the difference between the two groups was statistically significant(P < 0.05).The positive expression rate of TuM2-Pk was 35.71 % in? ~ ? stage and 77.27 % in ? ~ ? stage,the difference between the two groups was statistically significant(P<0.05).The positive expression rate of TuM2-PK was 33.33% in well-differentiated laryngeal carcinoma and 80.95% in moderate and poor differentiated laryngeal carcinoma,the difference between the two groups was statistically significant(P < 0.05).The positive expression rate of TuM2-PK was 91.67 % in tumour tissues with lymph node metastasis and 45.83% in tumour tissues without lymph node metastasis,the difference between the two groups was statistically significant(P<0.05).The positive expression rate of TuM2-PK in tumour tissue was 52.94% in the patients less than 60 years old and 68.42% in the patients not less than 60 years old,which were no significant differences between the two groups(P>0.05).The positive expression rate of TuM2-PK was 68.18% in persons who smoked no less than twenty cigrattes per day and 50.00% in persons who smoked less than twenty cigrattes per day,which were no significant differences between the two groups(P>0.05).2 The positive expression rate of PFKFB3 in laryngeal carcinoma tissue and adjacent mucosa was 52.78% and 20.00% respectively,the difference between the two groups was statistically significant(P < 0.05).The positive expression rate of PFKFB3 was 26.67% in well-differentiated laryngeal carcinoma and 71.43% in moderate and poor differentiated laryngeal carcinoma,the difference between the two groups was statistically significant(P<0.05).The positive expression rate of PFKFB3 was 83.33% in tumour tissues with lymph node metastasis and 37.50% in tumour tissues without lymph node metastasis,the difference between the two groups was statistically significant(P < 0.05).The positive expression rate of PFKFB3 was 42.86% in?~? stage and 59.09% in ?~? stage,which were no significant differences between the two groups(P > 0.05).The positive expression rate of PFKFB3 in tumour tissue was 47.06% in the patients less than 60 years old and 57.89% in the patients not less than 60 years old,which were no significant differences between the two groups(P>0.05).The positive expression rate of PFKFB3 was 59.09% in persons who smoked no less than twenty cigrattes per day and 42.86% in persons who smoked less than twenty cigrattes per day,which were no significant differences between the two groups(P>0.05).3 The positive expression rate of TKTL1 in laryngeal carcinoma tissue and adjacent mucosa was 58.33% and 15.00% respectively,the difference between the two groups was statistically significant(P < 0.05).The positive expression rate of TKTL1 was 35.71% in?~? stage and 72.73% in ? ~ ? stage,the difference between the two groups was statistically significant(P<0.05).The positive expression rate of TKTL1 was 33.33% in well-differentiated laryngeal carcinoma and 76.19% in moderate and poor differentiated laryngeal carcinoma,the difference between the two groups was statistically significant(P<0.05).The positive expression rate of TKTL1 was 83.33% in tumour tissues with lymph node metastasis and 45.83% in tumour tissues without lymph node metastasis,the difference between the two groups was statistically significant(P < 0.05).The positive expression rate of TKTL1 in tumour tissue was 52.94% in the patients less than 60 years old and 63.16% in the patients not less than 60 years old,which were no significant differences between the two groups(P>0.05).The positive expression rate of TKTL1 was 68.18% in persons who smoked no less than twenty cigrattes per day and 42.86% in persons who smoked less than twenty cigrattes per day,which were no significant differences between the two groups(P>0.05).4 The spearman's Rank Correlation Analysis showed that the positive expression of TuM2-PK and TKTL1 in laryngeal carcinoma had significant correlation(r=0.366,P < 0.05);the positive expression of PFKFB3 and TKTL1 in laryngeal carcinoma had significant correlation(r=0.442,P<0.05);But it had no significant correlation between TuM2-PK and PFKFB3 expression(r=0.044,P>0.05).Conclusions:1 The expression of TuM2-PK?PFKFB3 and TKTL1 were significantly higher in laryngeal carcinoma than that in adjacent mucosa.The expression of TuM2-PK?PFKFB3 and TKTL1 were related to lymphatic metastasis and the degree of pathological differentiation.The expression of TuM2-PK and TKTL1 were associated with clinical TNM stages.,while PFKFB3 was not.The three proteins were not significantly correlated with age and smoking status of patients.It suggested that these three proteins may have close relationship with the occurrence,invasion and metastasis of the laryngeal carcinoma.2 Both TuM2-PK and PFKFB3 expression were positively correlated with TKTL1 expression,suggesting that they may have a synergistic effect in the growth and proliferation of laryngeal carcinoma,and they may be expected to be clinical targets for laryngeal carcinoma treatment and offer theoretical evidence for the prognosis of the laryngeal carcinoma.
