| Objective:To observe the effects of different concentrations of Xiangxi Longshan Lilium extract on Mouse B-16 cells in cell viability, melanin content and tyrosinase activity. Methods:Select the same passage B-16 mouse melanoma cells and divided them in different groups, then lilium extract and vitamin C were added(vitamin C solution was diluted to the concentration of 0.01,0.02,0.04,0.08 g / L, vitamin C content was diluted into four different concentration according to the vitamin C content of lilium extract which was measured before) after 3 days affect of two solutions, B-16 cell viability was measured by MTT; B-16 Determination of tyrosinase activity in cells was measured by using Maeda’s method; and the measurement of the content of B-16 melanoma cells was conducted by Victoria’s method. Results: 1. MTT method of B-16 cell viability The inhibition effect of Xiangxi Longshan Lilium extract, vitamin C on mouse cells in cell viability increasing with concentration increases, indicating the cytotoxic effect of lilium extract is stronger than vitamin C. the concentration difference between the two groups was statistically significant(P <0.01), the comparison between the two groups was statistically significant(P <0.05). 2. Maeda’s method of measuring the activity of tyrosinase The effect of Xiangxi Longshan Lilium extract, vitamin C on tyrosinase activity is in a concentrationdependent inhibition. the concentration difference between the two groups was statistically significant(P <0.01), and the inhibition effect on tyrosinase activity of lilium extract is stronger than vitamin C, the comparison between the two groups was statistically significant(P <0.05). 3. Victoria’s method of measuring melanin content The inhibition rate of concentration of melanin in cells caused by Xiangxi Longshan Lilium extract and vitamin C increasing with the increase of the concentration. the inhibition rate of concentration of melanin in cells caused by lilium extract is better than vitamin C, the maximum rate of cell synthesis inhibition was gained when the lilium extract concentration is 0.08 g / L, the concentration difference between the two groups was statistically significant(P <0.01), the comparison between the two groups was statistically significant at the same concentration(P <0.05). Conclusion: the inhibition effect of lilium extract on cell viability, tyrosinase activity and concentration of melanin in cells is stronger than vitamin C, and the inhibition effect is concentration-related; But the whitening ingredients, whitening mechanism and cytotoxic ingredients of lilium extract are still unclear, further experimental study is needed. |
