Objective:Through the intervention of drug in asthmatic rats with Yang He Asthma-Easing Granules, to observe the effect of it on the subcutaneous fibrosis in the airway,and further discuss the intervention mechanism on EGFR pathways.Methods:Fifty SD rats of clean and healthy grade,were randomly assigned to five groups,with 10 rats in each group, for normal group, model group, and low-,and high-dose of Yang He Asthma-Easing Granules group,dexamethasone group. in addition to the normal group, the other 4 groups of rats were sensitized and excited by ovalbimin,successfully completed the model of asthma rats.Then,respectively,in the middle and Western medicine for eight weeks. After staining the pathological sections with HE staining method, to observe and evaluate the changes of pathological morphology. The expression levels of MMP-2 and MMP-9 in bronchoalveolar lavage fluid(BALF) were detected by enzyme linked immunosorbent assay(ELISA).The expression of airway subepithelial collagen deposition was measured by masson trichrome coloration.The expression of ?-SMA and FN was detected by immunohistochemistry. Protein Western blot(Western blot) method was used to detect the expression of EGF and EGFR protein in lung tissue. The expression of EGFR and m RNA in lung tissue was detected by real-time fluorescence quantitative reverse transcription polymerase(RT-PCR) method.Results:1 Compared with the normal group, around the airway and lung tissue of rats in model group,can be observed more inflammatory cells infiltration, addicted to the eosinophile cells seen, epithelial cell shedding, airway wall and smooth muscle thickening, lumen narrowing contains mucous substance; model group compared,Asthma-Easing Granules groups and dexamethasone group, inflammatory cell infiltration in the airway and lung tissue was significantly decreased, bronchial epithelial cell loss was relieved, and the mucosal layer was completely repaired.2 Compared with the normal group,The levels of MMP-2 and MMP-9 were significantly increased in the bronchoalveolar lavage fluid of asthmatic model group(P<0.01); Compared with the model group, the Yang He Asthma-Easing Granules group in high, low dose group and dexamethasone group expression levels all decreased(P<0.01); Compared with dexamethasone group, the effect of Yang He Asthma-Easing granules low-dose group expression levels were higher(P<0.05), the difference has statistical significance, high dose and no difference(P<0.05),.3 Compared with the normal group, the model group had a large amount of collagen deposition in the airway, and the value was increased obviously after the quantification of the fixed value(P < 0.01), the positive expression of-SMA and FN in the peripheral lung tissue was also significantly increased(P<0.01); Compared with the model group,the Yang He Asthma-Easing Granules group in high, low dose group, dexamethasone group of three groups of collagen deposition, a-SMA and FN positive expression were significantly reduced(P<0.01); Compared with dexamethasone group, effect of Yanghe Pingchuan granules low-dose group above three indexes were higher(P< 0.01), high dose and no difference(P > 0.05).4 Compared with the normal group, the expression of EGF, EGFR protein and mRNA in lung tissue was significantly increased in model group(P<0.01); Compared with the model,the Yang He Asthma-Easing Granules group in high, low dose group,dexamethasone group three groups of EGF and EGFR protein and m RNA in lung tissue of expression have decreased(P<0.01);Compared with the dexamethasone group, the protein expression of EGF and EGFR was higher in the low dose group(P<0.01), and there was no difference in the high dose group(P > 0.05). EGF m RNA expression, the high group and low group were significantly higher(P<0.01), the expression of EGF m RNA and low dose group of higher(P<0.05),high dosage group was no difference(P > 0.05).Conclusions:1 Yang He Asthma-Easing Granules group can reduce asthma rats lung tissue and peripheral airways to eosinophil eosinophil inflammatory infiltration, repair mucosal injury, so as to delay the airway wall and smooth muscle hyperplasia.2 Effect of Yang He Asthma-Easing Granules can decrease MMP-9 asthma MMP-2 in bronchoalveolar lavage fluid in expression, So as to reduce the chronic airway inflammation in asthmatic rats.3 Effect of Yang He Asthma-Easing Granules has the downregulation of expression?-SMA and FN, inhibitory effect of subepithelial collagen deposition. In order to relieve the asthmatic rat airway subepithelial fibrosis..4 Effect of Yang He Asthma-Easing Granules can significantly reduce the lung tissue EGF; EGFR expression, inhibit its activity, thus delaying the asthmatic rat airway subepithelial fibrosis.
