Effact Of Yanghe Asthma-easing Granules On Airway Mucus Hypersecretion Of Asthmatic Rats And TGF-β1/smad Signaling Pathways

Objective:Observing the intervention of YangHe Asthma-Easing Granules to airway mucushypersecretion of asthmatic rats,and further exploration its influence on TGF-β1/Smadpathways.Methods:Fifty SD rats were randomly divided into normal group, model group, andlow-,and high-dose of YangHe Asthma-Easing Granules group,dexamethasonegroup,each group of ten only.After successful induction of asthma with byovalbimin,and were given drug for treatment eight weeks.The changes of lung tissuepathology morphology was measured by HE.The expression of airway goblet cells wasmeasured by alcian blue/periodic acid Schiff(AB-PAS). The level of IL-4and IL-13inbronchoalveolar lavage fluid(BALF) was measured by enzyme-linked immuno sorbentassay(ELISA).The protein level of TGF-β1,Smad2and Smad3in rats lung tissue wasmeasured by Western blot.The gene level of TGF-β1,Smad2and Smad3in rats lungtissue and the gene level of Muc5ac in airway tissue was measured by real-time PCR.Results:1Asthma model group rats with a large number of inflammatory cells infiltration inthe lung tissue,trachea,mainly eosinophils,visize large amounts of mucus and gobletcell hyperplasia in the endotracheal.YangHe Asthma-Easing Granules groups,dexamethasone group around the bronchi and blood vessels in the lung tissue was notseen obviously inflammatory cell infiltration,bronchial mucosal was repaired morecompletely.2The levels of IL-4,IL-13in BALF of model group compared with normal groupsignificantly increased(P<0.01).The levels of YangHe Asthma-Easing Granulesgroups,dexamethasone group rats were obviously lower than model group (P<0.01);The level of dexamethasone group was significantly higher than that of low dose ofYangHe Asthma-Easing Granules group(P<0.01),but with high dose of YangHeAsthma-Easing Granules group no difference(P>0.05). 3The goblet cells in the airways of the asthma model group rats increased significantlycompared with normal group,the expression of airway tissue Muc5acmRNA washigher than normal group(P<0.01);The airway goblet cell hyperplasia in high and lowdose of YangHe Asthma-Easing Granules groups,dexamethasone group compared withmodel group significantly reduced(P<0.01);Expression of Muc5acmRNA in airwaytissue in dexamethasone group rats was lower than YangHe Asthma-Easing Granulesgroups (P<0.05).4The protein levels of TGF-β1,Smad2and Smad3and expressions of TGF-β1mRNA,Smad2mRNA and Smad3mRNA in lung tissue in model group significantly increasedcompared with normal group(P<0.01);Expressions in high and low dose of YangHeAsthma-Easing Granules groups and dexamethasone group was lower than modelgroup (P<0.01);Conclusions:1YangHe Asthma-Easing Granules can reduce the large number of inflammatory cellinfiltration,inhibit airway mucus plug formation,reduce airway mucosa edema andinflammation of the airway.2YangHe Asthma-Easing Granules can reduce the level of IL-4,IL-13in BALF ofasthma rats,improve airway inflammation of asthma rats.3YangHe Asthma-Easing Granules can inhibit goblet cell hyperplasia,reduce theexpression of Muc5acmRNA,thus may improve airway mucus hypersecretion ofasthmatic rats.4YangHe Asthma-Easing Granules can control airway mucus hypersecretion ofasthmatic rats through the TGF-β1/Smad pathways.
