Objective:The present study was using the miRNA chip to analysis difference of miRNA profiling between skin tissues from psoriasis vulgaris and normal people.After that we would predict the target gene,their function and how to paticipate in the psoriasis occurrence.Through the research above,we would further elaborate the mechanisms for pathogenesis,and provided a targeting therapy for clinical treatment of psoriasis.Methods:In the study,we chose 3 patients with vulgaris psoriasis and 3 normal person as control,then removed a little tissue from each person,and extracted and evaluated the qulity and purity of total RNA which must be consistent with request of the technology standard of Affymetrix microRNA chip,then fluorescent labeling,hybridization,the hybridized chips were scanned by Affymetrix GeneChip Scanner 3000,then used Affymetrix GeneChip Operating Software Versionl.4 to read and make data processing.By according the q-value(%)≤5 and fold chang≥2 we Chose the different genes and used the Human Target scan5.1 database and MAS system including Pathway and Go databases for the differential expression of miRNA target gene prediction and functional analysis,in order to build and pathological manifestations of psoriasis contact.Results:①There were 5 microRNAs different expressed in vulgaris psoriasis and normol people,Hsa-miR-1308 was upregulated expressed,downregulation of the remaining 4 including Hsa-miR-27a,Hsa-miR-199a-3p,Hsa-miR-199b-3p,and Hsa-miR-181a.The nucleotide sequence of these miRNA was focus on between 18 to 23 nucleotides.②Predicting target genes by Human target scan 5.1 database,we can get 2126 genes which were all negative regulated by the different miRNA.Then we used the Pathway and GO classification database which were belong to MAS systerm to anylysis the function of 2168 genes.The results showed that the target genes mainly invovled the participation of multiple signaling pathways,cell proliferation and differentiation,immune cell proliferation and regulation of biological processes.Conclusion:Psoriasis is a disease of genetic abnormalities,the miRNA can regulate a large number of mRNA negativly on the level of transcription,the regulated mRNA are interacted between each other,and fornat a network of co-regulation of the structure by a variety of ways to participate in the occurrence of psoriasis.
