MiRNA And MRNA Expression In Early Stage Of Skin Expansion And Key MiRNA Screening

Objective To establish early stage model of skin expansion; to determine the miRNA and mRNA expression in early stage of skin expansion.Method early skin expansion model was established using 6-8 weeks SD rat. Expanded skin samples were taken in the early stage and unexpanded skin was used as control. Affymetrix microRNA 3.0 and Affymetrix GeneChip Rat Gene 1.0 ST were undertaken to reveal the miRNA and mRNA expression change during early expansion stage. Statistic analysis was applied to filtrate differentially expressed miRNA and mRNA, foldchange of miRNAs was also calculated to determine key miRNAs. TargetScan, miRDB and IPA were applied to predict targets of these key miRNAs. Negative correlation between miRNA and mRNA was used to determine the real target of key miRNAs in the early stage of expansion.Result 11 key miRNA was detected and corresponding relation between miRNAs and their targets was also determined. These key miRNAs regulate pathways like TGF-? Signaling?Apoptosis Signaling and PDGF Signaling.Conclusion Present study determined 11 key miRNAs in the early stage of skin expansion and they regulate pathways play an important role in skin expansion. These key miRNAs could serve as targets to improve expansion efficiency and regeneration ability of skin.
