Objective:Discuss the association of Chlamydia trachomatis infection in the ovarian tissue and IL-12 gene polymorphism with epithelial ovarian cancer of China Hunan population.Method:There were 92 ovarian tissue of affecting epithelial ovarian cancer (EOC) cases being selected as case group, and 38 normal ovarian tissue cases as control group, the genome DNA of all subjects were extracted from ovarian tissue. All specimens had been carried out CT-DNA detection by PCR and IL-12 gene polymorphisms of all specimens were classified by PCR-RFLP technique, their results were assessed by statistical analysis. The epidemiological investigation had been undergone over all subjects with the relevant corresponding factors.Results:There were significant positive rate differences existing both in CT-16S DNA and CT-PD DNA over two groups. The distribution frequency of GG, AG and AA genotypes of IL-12A rs568408 gene differed in case group and control group (P=0.05). The frequency of GG genotype and non-GG genotypes distributed variously between two groups (P<0.05). No frequency variation existed in G/A allele (P>0.05). Frequency of AA, AC and CC genotypes of IL-12B rs3212227 indicated little difference over two groups (P=0.086), and frequency distribution varied between AA genotype and non-AA genotypes (P<0.05). It showed variation in frequency distribution of IL-12B rs32122227 A/C allele between two groups (P<0.05). The carriers of genovariation at both loci had 8.67-fold increased risk of epithelial ovarian cancer, compared with those carrying both wild-type genotypes. It indicated significant difference of frequency distribution over the genovariation at both loci of IL-12A and IL-12B and other genotypes in two groups (P<0.05), The logistic regression analysis showed that distribution of differences in the two groups was significant with respect to the abortion number of them, P for 0.009, however, other factors in the two groups showed little difference in frequency distribution, where P>0.05. The stratified analyses suggested that the genovariation at both loci has more effects on the following aspects:age (<50), age at menarche (<15), premenopausal, parities (>2 times), ligated contraception and artificial abortions.Conclusion:The present research shows that CT infection may promote the occurrence of epithelial ovarian cancer. IL-12B rs3212227 A/C allele variation may link to the susceptibility of epithelial ovarian cancer, at the moment, no evidence supports that the IL-12A rs568408 G/A allele variation with EOC. The genovariation of IL-12A and IL-12B may individually or jointly contribute to the risk of epithelial ovarian cancer. Epidemiological survey studies have indicated that artificial abortion is epithelial ovarian cancer risk factor. Among those are less than 50 and menarche age are less than 15, premenopausal, paries>2, ligated contraception and artificial abortions above, variation of two genes at both loci may contribute more to the occurrence of epithelial ovarian cancer, compared with other genotypes.
