Effect Of Zhi-chang-fa To Secretory Mechanism Of Neuropeptide SP、 CGRP、VIP Of Pulmonary And Intestinal Tissue Of Rats Combine Intestinalflora’s Imbalance With Allergic Asthma

Objective:Establishing the animal model of imbalance of intestinal flora combine with Allergic asthma, and treating it with "Zengye Chengqi Decoction". Through the observation of animal model of lung and intestinal’s pathological changes, pulmonary function, intestinal flora’s classification and count,EOS’s count of BALF and serum,lung and intestine’s neuropeptide,Understanding the therapeutic effect and secretion and regulation mechanism of neuropeptides of zhi-chang-fa on the animal model(imbalance of intestinal flora combine with allergic asthma).Method:Establishing the animal model of imbalance of intestinal flora,by drinking water solution of cefoperazone and gavaging with Candida albicans,sensitizing rats by intraperitoneal injection of gel sensitizer,it is formulated from OVA and aluminum hydroxide,establishing the animal model of allergic asthma,by inhalating of OVA in order to ultrasonic atomizing,taking"Zengye Chengqi Decoction"as the representative prescription on zhi-chang-fa.testing pulmonary’s function,and counting intestinal microflora,BALF’s cell and blood smear, removed pulmonary tissue and intestinal tissue for pathological examination and immunohistochemical examination of neuropeptide include SP,CGRP,VIP.Results:After oral administration,the rats of high-dose group of "Zengye Chengqi Decoction" and positive group, their asthma symptoms have improved significantly,high-dose group of Zengye Chengqi Decoction" can significantly improve the morphology of lung tissue and intestinal tissue,compared with the pathological model group,the group of "Zengye Chengqi Decoction" has manifest difference(P<0.05)on SP,CGRP,VIP of lung tissue and intestinal tissue,compared with medium-dose group and low-dose group,the high-dose group has manifest difference too.Conclusion:zhi-chang-fa has a good therapeutic effect on imbalance of intestinal flora with allergic asthma,especially on the imbalance of intestinal flora,verify the chinese medicine’s theory of "Lung and Large Intestine are interiorly-exteriorly related"through the mechanism of secret and regulate neuropeptide,it provides laboratory evidence for using zhi-chang-fa on pathological changes of Lung and Large Intestine (imbalance of intestinal flora combine with allergic asthma).
