Objective:Cervical cancer is the common disease of women malignant tumor,it's a serious threat to women's life and health. As a developing country, the new case rate of cervical cancer of china is 30% of the world.Cervical cancer patients have been growing at 2% to 3% a year.And in recent years, the majority of cervical cancer census showed the onset age tend to be younger.Now there are a large number of studies have shown that HPV infection, especially the high-risk type HPV infection is the vast majority of the main cause of cervical precancerous lesions and cervical cancer.And HPV16 and HPV18 are the most common types. PcG gene (Polycomb group) is a new discovered cancer-related gene in recent years.Emerging evidence suggests that PcG plays an important role in the stability of homologous abnormity gene in Drosophila development.It plays an critical role in embryonic development,tumor occurrence, tumor metastasis and maintaining stem cells.Now a number of studies have shown that PcG is a oncogene, participating in nasopharyngeal carcinoma, breast cancer, lung cancer, bladder cancer and many other kinds of human tumor occurrence and development.HPC2 is a member of the PcG gene family, and it is tested out that there is a lot of expression in a variety of cancers.The existing studies have confirmed that the PcG protein HPC2 can induce cancer cell apoptosis, regulate cell growth, etc.It also had been confirmed that HPC2 is overexpression incervical cancers Compared with the normal cervical tissue,however the mechanism of HPC2 protein in cervical cancer is still unknown, needing further study.Further understanding and researching of HPC2 bioinformatics functions and its relationships with cervical cancer are needed,aiming to provide a new direction for the research of the pathogenesis of cervical cancer occurrence.In this experiment,use methods of bioinformatics, analyzing the physical and chemical properties,hydrophilic/ hydroph obic,location in cancer cells,conserved domain of the HPC2 protein amino acid sequence, and carry on its system evolution relationship prediction.Aim to provide theoretical basis for the coming conduction of HPC2 related research.Afterwards we culture cervical carcinoma cells infected by different HPVs in vitro(Siha cell is infected by HPV16,Hela cell is infected by HPV18,C-33a cell is uninfected.).To explore HPC2 PcG protein expression in those three kinds of cervical cancer cells;And to compare the expression of HPC2 protein in different cervical cancer cells infected by different HPVs. Try to provide a new research direction of the mechanism of cervical cancer caused by HPVs. Methods:1. Culture three kinds of cervical cancer cell lines in vitro.2.Use fluorescence quantitative RT-PCR to observe the expression of HPC2 gene in different cervical cancer cells infected by different HPVs,and to explore whether there are difference in C-33a?Hela?Siha.3.Use western blot technique to compare whether the expression in Siha and Hela and C-33a cells exist differences. Results:1.The real-time fluorescent quantitative RT-PCR result shows that there are HPC2 gene expression in those cervical cancer cells with or without HPV infection.That the expression of HPC2 gene in Hela,Siha cells is lower than in C-33a cells,P is less than 0.05,there is statistical significance.No obvious difference was found between Siha and Hela cells in the expression of HPC2 gene,P=0.318>0.05.2.Western Blot result shows that there is HPC2 protein expression in those three kind of cervical cancer cells,HPC2 protein expressions of Siha,Hela cells is significantly lower than the C-33a cells.Conclusion:1.There is PcG HPC2 protein expression in C-33a,Hela,Siha cells.2.HPC2 gene and protein in HPV-infected cervical cancer cells is lower than the C-33a cells without HPV infection.No obvious difference has found in Siha and Hela both infected with HPCVS.3.HPC2 is associated with HPV infection.
