Objective: 1.To investgate the pathogenesis of PTEN/PI3K/AKT/mTOR signaling pathways in extra nodal NK/T cell lymphoma,nasal type.2.To explore the prognostic significance of PTEN gene of the patients with extra nodal NK/T cell lymphoma.Methods: 1.Collecting the pathological specimens of the patients with nasal type NK/ T cell lymphoma from June 2010 to March 2015 in the Affiliated Hospital of Luzhou medical college as the experience group.Another 20 cases of reactive hyperplasia lymph nodes were selected as the control group.2.Detecting the expression of p-AKT?p-4EBP1?p-mTOR in the control group and experimental group by using immunohistochemical EnVision.3.Detecting the expression of PTEN of the control group and experimental group by using immunohistochemical EnVision.4.Collecting the clinical data of the experimental group,analyze the clinical characteristics and prognosis of it,and then explore the relationship between the PTEN gene and its clinical features.5.Survival analysis was carried out using Kaplan-Meier method,and the survival rate comparison was performed by Log-rank test.Results: 1.p-AKT expression percentage of the experimental group were 82.35%,while that of the control group were 25.00%(X2=21.11,P<0.005).The expression rates of p-4EBP1 in the experimental group and the control group were 84.31% and 35%(X2=16.77,P < 0.005).The expression rates of p-mTOR in the experimental group and the control group were 60.78% and 40%(X2=2.50,P < 0.25).2.The expression rates of PTEN gene in experimental group and control group were 39.21% and 85%.3.There is no statistically significant difference between the expression of PTEN gene and the patients' age(>60 years??60 years),sexuality(male/female),group(A group?B group),Lactic dehydrogenase(rise up?normal),physical fitness scores(<2 points??2 points),the number of external invasion(<2??2),the incidence of the disease(in the nasal cavity?out of the nasal cavity)(P>0.05).4.IPI score between 0-2 group,PTEN positive rate was 53.85%,while the positive rate of the group between 3-5 scores was 24%(X2=4.76,P < 0.05),there is a statistically significant difference between the two groups.5.The median survival time of the PTEN gene negative group by Kaplan-Meier was 23 months,and 95% confidence interval was 17-28 months.While the median survival time of the PTEN gene positive group was 31 months,and the 95% confidence interval was 28-34 months.The 1 year and 2 year survival rates of the patients with the PTEN gene negative and positive in lymph node NK/T cell lymphoma were 70.8%,47.5%,87.8%,80.2%.The survival time of the PTEN negative and positive patients were tested by Log-Rank,X2=4.204,p<0.05(table 3.1),the difference was statistically significant.Conclusion: 1.There is an activation of PTEN/PI3K/AKT/mTOR signaling pathways in NK/T lymphoma,maybe it is related to the occurrence and development of nasal type NK/T cell lymphoma.2.There was no significant correlation between the expression of PTEN gene in patients with extra nodal NK/T cell lymphoma and the patients' age,sexuality,group,LDH level,physical fitness scores,the number and position of the extranodal involvement.While there was a significant difference with the disease stage(I-II stage and III-IV stage)and IPI scores(0-2 points,3-5 points).3.The 1 year and 2 year survival rate of the patients with PTEN negative NK/T cell lymphoma were significantly lower than those of the PTEN gene positive patients,and the difference was statistically significant.
