Objective:By studying effects of Fufang Xueshuantong Capsule on Bcl-2 and Bax in diabetic mice's retina, To explore the mechanism invigorating the circulation of supplement qi and nourishing Yin method in the treatment of diabetic retinopathy.Methods:Select 102 male SPF mice, after 10 hours fasting,91 mice were injected into STZ buffer (50mg/Kg) to settle DM mice's model, continuous injection once a day, for 4 days. Then use the high sugar and high fat feeding mice for 4 months, monitoring the blood sugar, DR mice's model was established gradually, then according to the weight and blood sugar levels stratified randomly divided into model group, high, medium and low dosages of Fufang Xueshuantong Capsule group, Qiming granules group and control group. Executed after 4 weeks corresponding drug lavage, apply retinal slices, immunohistochemical staining, the Image-Pro Plus6.0 Image analysis system to determine the average optical density of retinal tissue, detecting the expression of anti-apoptotic factor Bcl-2 and apoptotic factor Bax.Results: ?After 4 weeks treatment, the Bcl-2 average optical density in mice's retina of control group, model group, low, medium and high dosages of Fufang Xueshuantong Capsule group and Qiming granules group were 0.2068±0.0117? 0.1810±0.0125?0.1926±0.0160?0.1962±0.0184?0.1960±0.0152?0.2096±0.0156. Compared with control group, the expression of Bcl-2 average optical density in mice's retina of model group was much lower (P< 0.01); Compared with model group, the expression of Bcl-2 average optical density in mice's retina of Qiming granules group was much higher (P< 0.01); Compared with Qiming granules group, the expression of Bcl-2 average optical density in mice's retina of low dosages of Fufang Xueshuantong Capsule group was much lower (P< 0.05). Compared with model group, the expression of Bcl-2 average optical density in mice's retina of all Fufang Xueshuantong Capsule groups showed an increasing trend, but there was no statistical significance (P>0.05) ?The Bax average optical density in mice's retina of control group, model group, low, medium and high dosages of Fufang Xueshuantong Capsule group and Qiming granules group were 0.1003±0.0088? 0.1339±0.0121?0.1529±0.0369?0.1101±0.0258?0.0935±0.0213?0.1161±0.0292. Compared with control group, the expression of Bax average optical density in mice's retina of model group was much higher (P< 0.01); Compared with model group, the expression of Bax average optical density in mice's retina of high dosages of Fufang Xueshuantong Capsule group was much lower (P< 0.01); Compared with medium, high dosages of Fufang Xueshuantong Capsule group and Qiming granules group, the expression of Bax average optical density in mice's retina of low dosages of Fufang Xueshuantong Capsule group was much higher (P< 0.01). ?The Bcl-2 and Bax average optical density ratio in mice's retina of control group, model group, low, medium and high dosages of Fufang Xueshuantong Capsule group and Qiming granules group were 2.0758±0.2091?1.3626±0.1649?1.3221±0.3191? 1.8553±0.4122?2.1820±0.4666?1.9238±0.3675. Compared with control group, the expression of Bcl-2 and Bax average optical density ratio in mice's retina of model group was much lower (P< 0.01); Compared with model group, the expression of Bcl-2 and Bax average optical density ratio in mice's retina of medium, high dosages of Fufang Xueshuantong Capsule group and Qiming granules group was much higher (medium dosages of Fufang Xueshuantong Capsule group P< 0.05, high dosages of Fufang Xueshuantong Capsule group and Qiming granules group P< 0.01); Compared with medium, high dosages of Fufang Xueshuantong Capsule group and Qiming granules group, the expression of Bcl-2 and Bax average optical density ratio in mice's retina of low dosages of Fufang Xueshuantong Capsule group was much lower (P<0.01).Conclusion:Fufang Xueshuantong Capsule can increase the expression of anti-apoptotic factor Bcl-2, and reduce the expression of apoptotic factor Bax in C57 mice's retina, might be in the occurrence and development of diabetic retinopathy has certain inhibition, high, medium dosages of Fufang Xueshuantong Capsule group is significant.
