Study On Efficacy Of Fufang Xueshuantong Capsule In Antithrombus And Further On Its Mechanism

ObjectiveTo establish with rats the model of inferior vena cava thrombus,carotid arterio-venous bypass thrombus model and model of common carotid artery thrombus,and through which the antithrombotic effect of Fufang Xueshuantong Capsule was explored.And further,with the acute blood stasis rat model,the mechanism of antithrombotic effect of Fufang Xueshuantong Capsule was detected as well.MethodsThe rats were randomized into model group, Fufang Xueshuantong capsule at low, moderate, high dosages (0.375,0.75,1.5g-kg-1·d-1) group and Aspirin Enteric-coated Tablets (50mg-kg-1·d-1) group, each group was intragastric administration one time every day, consecutive7days. One hour after the last time of medicine, the vena cava inferior thrombus model and carotid arterio-venous bypass thrombus model were built, the effect of Fufang Xueshuantong capsule on wet weight and dry weight of thrombus, and thrombogenesis-inhibition rate in rats were observed. Be grouped according to above-mentioned method, established carotid artery thrombus model,meanwhile intragastric administration, consecutive5days. After the last time of medicine, the effect of Fufang Xueshuantong capsule on wet weight and thrombogenesis-inhibition rate in rats were observed. Add normal group on that basis of the first experiment, the rats were given subcutaneous injection of epinephrine, ice-bath and reducing feeding to establish acute blood stasis model. The effect of Fufang Xueshuantong capsule on Clotting time (CT), thrombin time (TT), and platelet aggregation function of rats was observed.One hour after the last administration, microcirculatory blood flow in rats’auricle was detected by laser doppler, the platelet aggregation by aggregometer,CT by slide method, plasma TT,PT,APTT,FIB,PLG,FDP by full-automatic coagulation analyzer and TXA2,PGI2,ET-1,CGRP of serum by ELIS A testing.Results.The results of study on prevention of inferior vena cava thrombus showed that compared with normal control group, a evident decrease in wet weight and dry weight of thrombus in rats of high and middle dosage groups of Fufang Xueshuantong Capsule was observed. With the carotid arterio-venous bypass thrombus model, a obvious decrease in wet weight and dry weight of thrombus in rats of high and middle dosage groups of Fufang Xueshuantong Capsule was showed as well.And with model of common carotid artery thrombus,a significant decrease in wet weight of thrombus in rats of all three dosage groups of Fufang Xueshuantong Capsule was detected.The results of the study on the mechanism of antithrombotic effect of Fufang Xueshuantong Capsule showed that,with the acute blood stasis rat model,compared with normal control group,the decline in rats’auricle microcirculatory blood flow,enhancement in platelet aggregation,evidently shortened CT,TT,APPT,PT, significantly raised FIB and remarkable descent in PLG were observed in model group.And,relative to normal control group,the evidently increased serum TXA2, obviously decreased serum PGI2the signicant increase in proportion of TXA and PGI2, evidently raised serum ET-1and remarkably declined serum CGRP were detected.Relative to model group,evident improvement for rats’auricle microcirculatory blood flow and platelet aggregation and remarkbly prolonged CT in all three dosage groups of Fufang Xueshuantong Capsule were observed;the evidently prolonged plasma TT showed in low and middle dosage groups of Fufang Xueshuantong Capsule;the significantly prolonged plasma APTT exhibited in middle dosage group of Fufang Xueshuantong Capsule; low and middle dosage groups of Fufang Xueshuantong Capsule showed a obviously raised plasma PLG;middle dosage group of Fufang Xueshuantong Capsule exhibited a remarkable decrease in plasma FDP;and the significant descent in serum TXA2was observed in low dosage groups of Fufang Xueshuantong Capsule; obviously increased serum PGI2and TXA2/PGI2were detected in all three dosage groups of Fufang Xueshuantong Capsule; serum ET-1showed a significant decline in low and middle dosage groups of Fufang Xueshuantong Capsule; remarkble raise in serum CGRP in low and high dosage groups of Fufang Xueshuantong Capsule was observed.ConclusionWith the model of inferior vena cava thrombus,carotid arterio-venous bypass thrombus model,the decrease in wet weight and dry weight of thrombus in rats all three dosage groups of Fufang Xueshuantong Capsule relative to those of model group. With model of common carotid artery thrombus,evident decrease in weight of thrombus in rats of all three dosage groups of Fufang Xueshuantong Capsule was detected.The result indicate that three model was successfully built,and each dosage of Fufang Xueshuantong Capsule showed the different efficacy in antithrombosis.In the further mechanism study,compared with model group,a raise in rats’auricle microcirculatory blood flow,CT, APTT,and TT, PLG PGI2and CGRP in plasm,and a decline in platelet aggregationFIB,FDP in plasm, TXA2, TXA2/PGI ET-1of serum were observed.In conclusion, Fufang Xueshuantong Capsule exhibited a antithrombus effect,which was caused possibly through the improvement in functions of coagulation, fibrinolytic system, platelet aggregation and endothelial cells.
