Objective To evaluate vitro anti-tuberculosis performance of calcium sulfate/poly amino acids artificial release material compounded triple anti-tuberculosisdrugs HRZ.Methods Soak the calcium sulfate/poly amino acids artificial releasematerial compounded triple anti-tuberculosis drugs HRZ and the calcium sulfate/poly amino acids artificial release material uncompounded triple anti-tuberculosisdrugs HRZ in the SBF. Detect the concentration of their extracts obtained in4thW,8thW,12thW with HPLC. Three groups were divided in this experiment:experimental group (A) was the extracts of calcium sulfate/poly amino acidsartificial release material compounded triple anti-tuberculosis drugs HRZ in4thW,8thW,12thW, divided into A4W, A8W, A12Wthree sub-groups, each sub-group of20samples;experimental control group (B) was the extracts of calcium sulfate/polyamino acids artificial release material uncompounded triple anti-tuberculosisdrugs HRZ in4thW,8thW,12thW, divided into B4W, B8W, B12Wthree sub-groups,each sub-group of20samples; blank control group (C group) was distilled watergroup, divided into C4W, C8W, C12Wthree sub-groups, each sub-group of20samples. Three groups of samples and standard M. tuberculosis H37Rv were addedinto BacT/ALERT3D co-culture fast training instrument and L-J medium, observethe positive rate of culture results and statistical analysis.Results The result of the extracts concentration of the calcium sulfate/poly amino acids artificial release material compounded triple anti-tuberculosis drugsHRZ(? g/ml): the concentration of INH in4w,8w,12w:142.65±2.23,47.83±2.91,0.68±0.17; the concentration of RFP in4w,8w:70.69±7.16,39.95±3.98,the concentration can't be detected in12w; the concentration of PZA in4w,8w,12w:893.56±7.09,229.87±9.56,6.36±0.57. After co-cultured in BacT/ALERT3D system, the positive rates of A, B, C three groups in4W,8W,12W were: Agroup of15%,25%,55%; B group of80%,85%,80%; C group of90%,95%,90%.There were significant differences (P<0.05) between corresponding samples ofgroup A and group B. There were no significant difference (P>0.05) betweensamples of group B and group C. After co-cultured in L-J medium, the positiverates of A, B, C three groups in4W,8W,12W were: A group of15%,35%,25%; Bgroup of80%,80%,85%; C group of85%,90%,95%. There were significantdifferences (P<0.05) between corresponding samples of group A and group B.There were no significant difference (P>0.05) between samples of group B andgroup C.Conclusions (1) The calcium sulfate/poly amino acids artificial releasematerial compounded triple anti-tuberculosis drugs HRZ has a very nice vitroanti-TB effect till the84thday.(2) The bactericidal effect of the calcium sulfate/poly amino acids artificial release material compounded triple anti-tuberculosisdrugs HRZ reduced with the extension of time.(3) The combination of BacT/ALERT3D system and L-J medium can improve the detection accuracy andprovide a good method for the laboratory.
