Influences Of Ozonized Saline On Behaviors Of Male Rat Models With Incisional Pain And Levels Of Plasma TNF-a And IL-6

Objectives:By observing changes in behavior, plasma TNF-a and IL-6levels after rat plantar incision, we investigate the analgesia effect and the possible function mechanism of intravenous administration with different concentrations of ozonized saline (OS).Methods:Fifty-four health adult male Sprague-Dawley rats were randomly divided into two groups.Group1:24rats were randomly divided into four groups,6in each group.The rats of group A were intravenously administered via the vein in the tail with5ml/kg oxygen saline,10min before the incision. The rats of group B were intravenously administered with20?g/mlOS5ml/kg. The rats of group C were intravenously administered with30?g/mlOS5ml/kg. The rats of group D were intravenously administered with40?g/mlOS5ml/kg.An1cm long incision was made in the right plantar surface from the heel to the toes according to the method described by Brennan under sevoflurane anesthesia.The50%paw withdrawal mechanical threshold(50%PWMT) and cumulative pain score(CPS) at the times of24h before and2,6,24,48h after surgery were underwent.Group2:30rats were randomly divided into five groups,6in each group.Group A,B,C,D processed as well as group1.All groups except group E needed to be made the model of incisional pain.The right ventricular blood2ml of the rat was taken out2h after the operation,then the levels of plasma TNF-a and IL-6were detected by using ELISA.The adverse effects were recorded during the treatment of OS.Results:(1)No chemical vasculitis, other adverse reactions and death were occurred in the rats treated solitarily with OS.(2)The50%PWMT in group B compared with group A were not obvious at each time point after surgery(P>0.05). The50%PWMT in group C and D were higher than those in group A at each time point after surgery(P<0.05).(3)The CPS in four groups were zero before surgery.The CPS in group B,C,D were lower than those in group A after surgery(P<0.01).The group D were lower than group C. The group C were lower than group B.(4)Compared with group E,2h after the operation, the levels of plasma TNF-a of group B and D were not obvious(P>0.05). The levels of plasma TNF-a of group C and A were higher than group E(P<0.05). The levels of plasma TNF-a of group C were higher than group A(P<0.05).(5) Compared with group E,2h after the operation, the levels of plasma IL-6of group A were not obvious(P>0.05). The levels of plasma IL-6of group B,C,D were higher than group E(P<0.05). The levels of plasma IL-6of group D were the highest.Conclusions:(1)Intravenous injection of ozonized saline can inhibit the incisional pain in rats.(2)The analgesia effect of ozonized saline was dose-dependent.(3)Intravenous injection of ozonized saline can increase the levels of plasma TNF-a and IL-6.(4) Intravenous injection of ozonized saline showed no significant adverse reactions in rats.
