Tomato(Solanum lycopersicum)is an important cash crop.Widely cultivated in China,it is loved by people because of its high nutritional value and high quality and low price.However,in the flowering period,the unsuitable environment,such as high temperature,low temperature,drought and so on,causes bad pollen pollination and falling fruit,which affects the yield of tomato.Pollen development is very important for the flowering plants of tomatoes.There are many factors affecting the development of pollen,such as environmental conditions,hormone levels,and so on.In recent years,with the continuous development of molecular biology,it is found that pollen development is a complex process controlled by a variety of genes.IDA(INFLORESCENCE DEFICIENT IN ABSCISSION)was first considered as a signal peptide regulating the abscission of Arabidopsis flower organs.The research group found that the regulation of SlIDA in tomato was not obvious in the earlier period.This study found that the expression of SlIDA in stamen was very high,and the expression of stamen and pollen in ProSlIDA:: GUS transgenic tomato was found by GUS staining.The function of SlIDA in tomato pollen development was analyzed based on the SlIDA RNAi silencing plant.The results are as follows:1.The temporal and spatial expression characteristics of tomato SlIDA gene showed that SlIDA gene was mainly expressed in stamens of tomato floral organs.2.Construction of ProSlIDA:: GUS transgenic tomato.The results of tissue and organ staining showed that SlIDA was mainly expressed in stamen pollen.3.Screening T5 generation SlIDA silenced transgenic plants.It was found that the seed setting rate and node rate of RNAi lines decreased significantly.Even a few RNAi strains were found to be deformed fruit with few seeds.This indicated that SlIDA affected seed setting and nodal rate.Hybridization results showed that RNAi plants mainly affect pollination and fertilization of pollen.4.SlIDA RNAi strain is pollen grain deformity,stamen without cracking,pollen viability and pollen germination rate are significantly lower than wild type,This suggests that SlIDA affects pollen development.5.NBT and DAB staining showed that the accumulation of stamen ROS decreased significantly during pollen development of SlIDA RNAi silencing plants.These results indicate that SlIDA can affect the fruit setting rate and seed setting rate of tomato and affect tomato yield through pollen development.
