NK cells,CD4~+T cells and CD8~+T cells secrete Interferon gamma when antigeninfects animal body and Interferon gamma is a homopolymer protein which is 34KD and it has extensive biological functions.In clinical practice,interferon gamma applies to antiviral treatment and antiviral treatment,unfortunately,it often accompanied by anorexia,frequent vomiting,diarrhea,slow emaciation and other digestive system side effects.So,it is very necessary to explore the regularities and mechanisms of i interferon gamma in regulating the digestive system.Interferon gamma receptor(IFNR-gamma)is the basis for conducting the physiological functions of interferon gamma on target cells.It is well known that the autonomic nervous system is one of the leading systems for regulating digestive system activity.In this study,immunofluorescence labeling technology was used to detect the distribution characteristics of Interferon gamma receptor and acetylcholine receptor(M3m Ach R)and adrenergic receptor(?3AR)on various layers of the duodenum of rabbits and BL-420F biological signal collection and analysis system was used to detect the synergistic regulative action and mechanism of interferon gamma and bethanechol chloride on duodenal motion.The major findings were as following:1?Immunofluorescence staining results:In the duodenum of domestic rabbits,the positive products of IFNR-gamma were mainly distributed in the outer membrane,submucosa and mucosa layer,Intrinsic layer of mucosa;A small number of positive products were found in the mucosal myometrium and myometrium.The immunoreactive products of M3m Ach R were mainly distributed in the muscular layer,and the smooth muscle of the outer longitudinal layer of the inner ring layer had bright green fluorescence,which was strongly positive.?3ARpositive product mainly distributed in the muscular layer and mucosa,muscularis smooth muscle,dark green fluorescence is weak positive,positive product distribution in the membrane layer in the connective tissue cells in the lamina propria and small lymphocytes,bright green fluorescence.2?The results of tests on duodenal myoelectric index with different concentrations of IFN-tests showed that:IFN-?have a slight promoting effect on duodenal index at low doses and a certain level of inhibition at high concentrations.3?The synergistic effects of interferon gamma and ammonium chloride methyl choline in duodenal myoelectric index showed that with the addition of acetylcholine,the effect of interferon gamma on duodenal myoelectric index has become inhibitory,and the inhibition is most obvious when the interferon gamma concentration is 250,000 IU/kg.Low interferon gamma conditions,different concentration of acetylcholine adjustment of myoelectricity index curve movement relatively consistent,when interferon gamma dose,myoelectricity under different concentration of acetylcholine adjustment index curve movement will show the relative complexity.The above results showed that:1)Morphological results revealed that The smooth muscle cells of duodenal muscle layer are not the direct targets of Interferon gamma.2)The effect of interferon gamma on duodenal myoelectric index indicates that interferon gamma have promote in low-dose and a influence of inhibition in high-dose on duodenum.The performance of interferon gamma and cholinergic nervous system in duodenal myoelectric index indicates that the inhibition of interferon gamma to duodenal myoelectric index is significantly enhanced in the presence of acetylcholine.It is speculated that the mechanism of interferon gamma's action on duodenal smooth muscle is indirect through the cholinergic nervous system.
