Soybean is an important grain crops and oil crops.In recent years,the main direction of research is to cultivate new soybean varieties with good traits or to improve the existing soybean varieties.EMS with high mutation frequency,wide range of mutation is random,so it is widely used in the construction of mutant population.According to reports,the existing research building the soybean mutant library by EMS mutagenesis,but how to choose whether the correlation with the best treatment methods and traits of mutant library of mutants of the results of the present study is less.This paper mainly studies the development of soybean and its resistance to Phytophthora root rot of mutant population analysis,results of the study are as follows:1.The experimental design of 6 EMS concentrations?0%,0.15%,by 0.3%,0.4%,0.5%,0.6%?and 4 mutation time?3 h,6 h,9 h,12 h?combination,based on the mutation after treatment of soybean YD,V15,LX,germination rate,germination rate decline,seedling emergence the rate and seedling rate statistics and analysis,to determine the soybean YD,V15,LX best mutagenic treatment conditions,namely EMS treatment concentration was0.35%,treatment time was 12 h.2.The processing conditions of soybean YD,V15 and LX with the best mutation treatment conditions,and the soybean YD,V15,LX in M2 groups were constructed.In the growth period of soybean YD,V15,LX in M2 group were observed.Results showed that there were Leaf Chlorosis,leaf curling,leaf color,leaf,leaf malformation,seed coat harmonia,shriveled seeds,dry seeds,small seeds,hilum color,single cotyledon mutant,double mutant dwarf stem,mutant,mutant plants growing slowly,100 grain weight,100grain weight decreased,plant height increased,plant height decreased.The total mutation frequency of soybean YD was 11.84%,the total mutation frequency of soybean V15 was13.05%,and the total mutation frequency of soybean LX was about 8.53%.3.With the known resistance of soybean YD and JKD as the control of the wild type soybean V15,YD,LX with the Phytophthora sojae.The results showed that soybean YD resistant varieties,soybean V15 resistant varieties of soybean LX for susceptible varieties.At the same time,soybean YD,V15,LX of M2 group were inoculated with Phytophthora sojae,and the mutant lines with increased resistance were successfully screened in the M2population of soybean LX-8?LX-50?LX-95?LX-67?LX-48?The results indicated that the mutant YD,V15 and LX mutants were successfully constructed by EMS mutagenesis,and the mutation types of YD,V15 and LX mutant libraries were analyzed.At the same time,the wild type soybean YD,V15 and LX resistance to Phytophthora sojae,the soybean YD,V15 and LX of the M2 group were inoculated with Phytophthora sojae,LX M2 in soybean group successfully screened resistant mutants increased.The study provided an important clue for the study on the mutation of soybean mutant library and the development of resistant varieties of Phytophthora sojae.
