| Gelsemium elegans Benth is a kind of evergreen vine belongs to the genus Gelsemium.In their knowledge,the plant has the effect of Anti-cancer and sedative hypnotic that make the plant popular among the field of medical.In addition,Gelsemium elegans Benth has shown the growth promoting and Anti-parasite effect on certain animals which made the Gelsemium elegans Benth reveals the application value of livestock breeding and poultry raising.Objective: This study investigated the effect of ethanol extract,acid water extract and water extract from Gelsemium elegans Benth on the motility of isolated intestine and situ gastrointestinal tract,and the effect of 3 extracts on proliferation of IPEC-1 cell line,By the method of molecular biology,animal Physiology,Pharmacology toxicology etc to ascertain the effect of 3 extracts of Gelsemium elegans Benth on the motility of gastrointestinal tract;To provide scientific basis for classifying the growth promoting mechanism of Gelsemium elegans Benth.Providing theories for the application and exploiting of Gelsemium extract preparation.Methods: The effect of Gelsemium extract on the contract of duodenum,jejunum,ileum respectively was determined by BL-420(F)biological function analysis system.Establishing the situ gastrointestinal tract model to classify the effect of 3 kinds of Gelsemium extracts on emptying of gastric contents and advancing function of the intestine.The proliferation of IPEC-1 cell line is established by WST-1.The effect on IPEC-1 cell cycle of 3 extracts is given in flow cytometry.Result: 3 kinds of Gelsemium extracts induced the concentration-dependent suppression in the contract strain of isolated intestinal smooth muscle,Showing the inhibition of intestinal motility.This inhibit effect is most remarkable in isolated jejunum.All the 3 kinds of Gelsemium extracts can remarkably inhibit the function of gastric emptying and intestinal propulsion on a mouse in a far less than their LD50 dose.All the 3 kinds of Gelsemium extracts can promote proliferation of IPEC-1 cell line.But the effect of promoting proliferation decreased progressively with increasing dose.Acid water extracts and water extracts of Gelsemium elegans Benth shorten the G1 PHase and extend the S PHase of cell circle in IPEC-1 cell line markedly.Ethanol extract of Gelsemium elegans Benth affect the cell circle was not noticeable.Conclusion: 3 kinds of Gelsemium extracts showed the inhibition of intestinal movement Inhibiting the function of gastric emptying and intestinal propulsion on mouse.Promoting proliferation of IPEC-1 cell line.Gelsemium extracts presented a potential value of developing into drugs regulating intestinal function. |
