In this study,the bacterial composition of tick’s body,salivary gland and saliva,which was obtained from partially or fully engorged adult Rhipicephalus microplus females,were analysed by PCR-DGGE and NGS.The result of PCR-DGGE revealed that partially or fully engorged tick’s body and saliva had 7 predominant bacteria,5 predominant bacteria,12 predominant bacteria and 14 predominant bacteria respectively.Among them,the Rickettsia peacockii,Rickettsia Heilongjiangensis,Rickettsia slovaca,Escherichia coli and Sphingomonas were shared between the body and saliva,but their abundances were diverse among samples extremely.In saliva,the fully engorged tick just more than partially engorged tick about two species of bacteria.They were affiliated with Moraxellaceae bacterium and Acinetobacter calculations.Bacteria’s abundance were diverse significantly in two stages of saliva.The result of NGS showed that partially or fully engorged tick’s body and saliva had 336 OTUs,334 OTUs,330 OTUs and 296 OTUs respectively.Among them,147 OTUs were shared between body and saliva.Except these OTUs,there were 67 OTUs,54 OTUs,66 OTUs and 43 OTUs specific to partially or fully engorged tick’s body and saliva respectively.The comparison of microbiome among the whole body,salivary gland and saliva show that partially engorged tick’s body,fully engorged tick’s body,partially engorged tick’s salivary gland,fully engorged tick’s salivary gland,partially engorged tick’s saliva and fully engorged tick’s saliva had 331 OTUs,313 OTUs,103 OTUs,202 OTUs,341 OTUs and 302 OTUs respectively.Among them,22 OTUs were shared between body,salivary gland and saliva at two stages.Except these OTUs,there were 67 OTUs,54 OTUs,22 OTUs,116 OTUs,66 OTUs and 42 OTUs specific to partially or fully engorged tick’s body,salivary gland and saliva respectively.This investigation revealed some pathogens such as Rickettsia,Coxiella,Propionibacterium acnes and Moraxella osloensis were abundant in Rhipicephalus microplus’ s saliva,implied these bacteria might transmit to human or animal by biting,developing multiple bacteria disease.
