Variety identification and genetic purity testing are two important issues in crop seed quality control. Basic principles and application of the genetic marker analysis technology. Such as morphological marker, isozyme,Seed storage protein, RFLP, RAPD, SSR, ISSR, AFLP in seed testing of crops are reviewed on this report; and assessed all the methods about purity testing objectively and justified. The identification of the genetic purity of F1 hybrid seeds in new variety "ShaanYou No8"was made via morphological of seedling, isozyme. Seed storage protein and RAPD ( Random Amplified polymorphic DNA) markers, achevied the following results: 1. using the morphological of seedling could identify the male line from the F1 generation of "ShaanYou No8"but couldn't identify the female line from the F1 generation, So. it is valueless in practice. 2. Application of EST-isozyme and POD-isozyme electrophoresis for purity testing and cultivar identification of F1 hybrids of ShaanYou No.8, the isozyme patterns showed, the Est-isozyme and POD-isozyme can only identify the male line from the F1 generation, but not identify the female line from the F1 generation(the isozyme patterns were identity completly in female line and F1 generation) so the EST isozyme and POD-isozyme can't identify the seed purity of "ShaanYou No.8"efficiency. 3. the seed storage protein patterns of "ShaanYou No.8"can identify male line. Female line and F1 generation each other. But because the seed of rapeseed (Brassica napes) is too small to extract the seed storage protein. So it can not do in practice. 4. The genomic DNAs extracted from yong levels of rapeseed hybrids "shaanyou No.8"and its parents were analyzed using polymerase chain reaction (PCR) with arbitrary 10-mer primers to generate random amplified polymorphic DNA(RAPD)markers.Primer BA208 could generate parental complementary characteristic bands:BA208-1050bp,BA208-1250bp, Primer BA497 could generate paternal characteristic band BA497-870bp.All the characteristic bands screened were found in "shaanyou No.8"hybrids。The"shaanyou No.8"can be identify from five rapeseed hybrids with BA497 ,The reliability of this result was confirmed by the phenotypes of hybrids seed lots in the field。Identification of the purity in F1 population using two primers was discussed in this paper。Using RAPD can identify the seed purity of "shaanyou No.8". |
