This experiment changed the nutrient composition of the diet and designed four different NDF levels,which were 32%,35%,38%,41%.at the same time,using four body weight similar?150+5?Kg Tarim Wapiti with permanent rumen fistula,designed by 4×4 Latin square.This study was conducted to the effects of dietary different NDF levels on rumen protozoa species,ruminal environment and rumen protozoa population structure of the Tarim Wapiti.Then results were compared between three kinds of stage.The first experiment,The results show:through morphological observation studies,the two catalogue of protozoa,such as Trichostomatida and Entodiniomorphoda.The three family of protozoa,sach as,Isotrichae,Belphaocorythidae and Ophryoscolidae.The three subfamily of protozoa,such as Entodiniinae,Diplodiniinae,Epidiniinae.The16 genus of protozoa,such as Isotricha,Dasytricha,Oligoisotricha,Charoina,Entodinium,Eodinium,Eremoplastron,Diplodinium,Eudiplodinium,Ostracodinium,Polyplastron,Metadinium,Enoploplastron,Diploplastron,Elytroplastron,Epidinium,etc,were found 176species of rumen protozoa.The second experiment analyzed the activity,quantity and frequency of protozoa in the rumen of Tarim wapiti using by feeding different dietary NDF levels,microscope and blood cell counting plate method.The results showed that,with increased NDF level in diet,The Rumen protozoa activity was continued to increase,and the total number of rumen protozoa was increased significantly.The dietary different NDF levels had significant effect on the frequency of Entodinium,Epidinium,Metadinium,Diplodinium,Isotricha,Dasytricha and Eudiplodinium?P<0.05?,but the three genus of protozoa frequency difference was not significant such as Enoploplastron,Eremoplastron,Elytroplastron?P>0.05?.High-throughput sequencing technology did not significantly affect the diversity and richness of protozoa in the rumen of red deer in NDF.According to the results of 18S rRNA gene comparison of rumen protozoa,the dietary different NDF levels had significant effect on the frequency of Entodinium,Polyplastron,Metadinium,Diplodinium,Diploplastron and Cycloposthium?P<0.05?,but the Isotricha frequency difference was not significant?P>0.05?.Under different NDF levels,the richness of the Entodinium and Polyplastron was higher than that of other genera.Through the study of morphological identification and high-throughput sequencing technology on rumen protozoa population structure comparison,the changing trends of Entodinium and Metadinium are consistent,but the changes in other genera are inconsistent.A preliminary study on the effect of NDF level on the population structure of rumen protozoa has important reference value for understanding the appropriate mechanism of Tarim Wapiti on low quality roughage.The third experiment,different dietary NDF levels was determinated on the ruminal environment,nutrient digestibility and rumen fiber degrading enzyme of the Tarim wapiti,The dietary different NDF levels was no significant effect on the pH value,Propionic acid,isobutyric acid,butyric acid,isovaleric acid,pentanoic acid,protozoan protein of Tarim Wapiti?P>0.05?.It was a significant effect on the NH3-N,acetic acid,Bacterial protein?P<0.05?.The apparent digestibility of the DM,CP,NDF was increased with the improvement of NDF levels,but the OM,ADF was decreased with the improvement of NDF levels.The Content and activity in the Xylanase,Cellobiase,CE,CMC was increased with the improvement of NDF levels?P<0.05?.
