| Tarim Wapiti deer has an unique enviroment,which mainly in area of Tarim River and Kongque River.Compared with other deers,Tarim Wapiti deer antler has faster growth and higher quantity. Tarim Wapiti deer antler of high yield was conducted to clone sequence of c-fos gene for the first time. It is very meaningful for further investigating molecular mechanism of c-fos.And providing the first materials to molecular marker-assisted selection to anter performance of Tarim Wapiti deer,we detected the c-fos gene polymorphism site and found its polymorphism assosiation with antler weight of Tarim Wapiti deer. The results were as follows:1.On the bovine c-fos gene(GenBank accession number:NM-182786.2) four exons separately designed four pairs of primer, a total of1952bp length of Wapiti deer c-fos cDNA sequence was successfully cloned. PCR products are596bp,441bp,742bp,704bp.Sequence alignment found that Tarim Wapiti deer c-fos the homologies of red deer c-fos with those of cattle, sheep, horsesã€human were97%,96%,94%,92%respctively.2.In sequence of Wapiti deer c-fos gene exon2,one SNP was found. Analyzed by PCR-SSCP,the results was follows:(1) There were three kinds of genetic type, respectively for the CC typeã€CT and TT type. Three genotypes CC,CT and TT was found and its genotype frequency was0.7045ã€0.0682ã€0.2273respectively.Gene frequency of Alleles of C and T were0.7386and0.2614.(2)By the chi-square test,Tarim Wapiti deer population were not in the state of Hardy-Weinberg(p<0.05).3. Assosiation analysis of Wapiti deer antler and genotype frequency, found that weight of Tarim Wapiti deer antler had no siginififcant effect of genotype(P>0.05). |
