| Spirotetramat is a new type of diketo acid pesticide,which is introduced by Bayer in 2007,has two-way internal absorption,and can effectively prevent and cure various kinds of sucking mouthparts pests,and the insecticidal effect can be achieved by inhibiting insect lipid synthesis.At present,there are few studies on the environmental toxicity of spirochthyl ethyl ester,especially on aquatic organisms.In this experiment,zebrafish embryos were taken as the research object,and the main contents of the study included the following aspects:(1)spirotetramat on teratogenic effects in zebrafish embryo development;(2)effects of spirotetramat on lysyl oxidase process in zebrafish embryo development;(3)effects of spirotetramat on oxidative stress system in zebrafish embryo development;(4)effects of spirotetramat on lipid synthesis pathway the process in the development of zebrafish embryo;(5)effects of spirotetramat on energy metabolism in zebrafish embryo development.The effects of spirochthyl ester on the embryonic development of zebrafish were analyzed.The results obtained from the experimental study on the embryonic development of zebrafish are as follows :(1)spirotetramat will lead to delay the development of zebrafish embryo,abnormal rate increased with increasing concentration,resulting in embryonic yolk sac edema,notochord flexion,pericardial edema,deformity type tail bending;(2)spirotetramat inhibits the expression of lysine oxidase gene in the pre 48 hpf stage of the embryo;(3)the activity and expression of superoxide dismutase(SOD)in 72 hpf were significantly inhibited by spirotetramat;the expression of glutathione peroxidase(GPX)was stimulated at 24 hpf stage,and was significantly inhibited at 48 hpf stageby spirotetramat;the expression of catalase(CAT)was inhibited at 72 hpf stageby spirotetramat;the malondialdehyde(MDA)concentration decreased significantly only at 72 hpf stage(2mg / L),but there was no significant change in other time and treatment groups;(4)spirotetramat significantly inhibited the activity and expression of target acetyl coenzyme A carboxylase,and inhibited the expression of fatty acid synthase and sterol regulatory element binding protein gene;(5)spirotetramat inhibited the expression of adenylate activated protein kinase(AMPK)at the 24hpf;the ratio of AMP/ATP was up-regulated and the activity of AMPK was increased in the pesticide treated group at 24 hpf stage.However,at 48 hpf returned to normal and inhibited the gene expression of ATPase at the first 24 h.The experimental results show that spirotetramat treatment may result in fetal growth retardation,tissue deformity;after treatment,the embryo development retardation and abnormal tissue development are related to the lipid synthesis and energy metabolism pathway in the development of zebrafish embryos;oxidative stress caused by occurrence of zebrafish embryos,but did not cause oxidative damage;during zebrafish embryonic development deformity is due to spinal cord spirotetramat after treatment resulted in decreased expression of lysyl oxidase caused. |
