Spirotetramat is a tetramic acid pesticide against sucking pest species, with both-way transmission, longer duration and remarkable prevention effects. Its up-to date aquatic toxicity had been rarely mentioned in literatures. In recent study, female zebrafish was used as research object.The effects of spirotetramat on oxidative stress system in ovary and liver were studied as well as the pesticide disrupt gonadotropic hormone of fish in 28-days’ toxicity tests.Zebrafish were exposed to spirotetramat at various concentrations(36, 360 and 720 μg/L),collect samples at the 14 th, 21 st and 28 th day. The activities of SOD and CAT, and the content of MDA, FSH, LH and VTG were determined by ELISA kits. The expression of genes were simultaneously detected with real-time quantitive PCR.The results indicated:(1)The spirotetramat caused the decrease of SOD activities in ovary. The ovary exposed to the 360 and 720 μg/L concentrations leaded lipid peroxidation and the content of MDA increase after 21 days. The m RNA levels of Cat and Cu/Zn-sod were significantly up-regulated after 14 and 21 days.(2)The activities of CAT and SOD, and the increase of MDA contents indicated the lipid peroxdation in the liver. The m RNA levels of Cat was increased after 14 and 21 days, then returned to normal levels after 28 days.(3)Exposed to spirotetramat at the concentration 720 μg/L 14 days caused the content of FSH decreased, but the content of VTG was increased. Meanwhile, the relative expression of FSHR was increased. Spirotetramat had no effect on the content of LH, but increased the relative expression of LHR.The results of assays in this study indicated the spirotetramat cuased the oxidative stress in zebrafih ovary and liver. Effect on the activities or expressions of enzymes and caused the content of MDA accumulated, and had the risk of potential oxidative damage in organism. Spirotetramat influences the content of FSH and VTG and the relative expression of LHR was increased, it means that the pesticide had endocrine disruptor activity in the female zebrafish.
