Triplophysa pappenheimi belongs to Cobitidae,Nemacheilinae and Triplophysa.In this study,the Yellow River basin of west China different waters of Triplophysa pappenheimi were collected from different geographical populations,Using the mitochondrial DNA control region(D-loop)sequence and the cytochrome b gene(Cytb)sequence,the genetic diversity of the loach population of different habitats or the Yellow River Plateau was analyzed,and the genetic composition and distribution of the loach populations in the Yellow River Plateau were discussed.st.then,Extracted and determinated the whole sequence of Triplophysa scleroptera.comparing with the complete mitochondrial genome of Triplophysa pappenheimi which included in NCBI,Analysised the structure characteristics,to enrich the genetic data of Triplophysa pappenheimi.Meanwhile,It provides the theory basis for effective protection and Reasonable use of Triplophysa pappenheimi.The main results are as follows:(1)The Cytb gene sequences of different waters of Triplophysa pappenheimi population has been analysised,Research different waters of Triplophysa pappenheimi‘s genetic and variation.The results show: The total length of Triplophysa pappenheimi Cyt b gene was 1140 bp(including two deletion sites),and 52 polymorphic loci were detected.There were 20 simple information loci and 37 haplotypes,5 groups of base content is almost the same,T,C,A,G were 32.3%,25.4%,26%,16.3%.Showing a higher haplotype diversity with lower nucleotide polymorphism Pattern.(2)Mitochondrial DNA sequence control of different waters of Triplophysa pappenheimi population has been analysised,Research different waters of Triplophysa pappenheimi ‘s genetic and variation.The results show : The total length of Triplophysa pappenheimi D-loop gene was 943 bp(no deletion).Among which there were 30 polymorphic loci,18 simple information loci and 33 haplotypes.The average content of Ctyb gene was 32.7%,21.6,32.3% and 13.4%,Consistent with Cytb,the same haplotype diversity and lower nucleotide diversity patterns were also observed.(3)The Cytb gene and D-loop region of different populations were analyzed by constructing the corresponding phylogenetic NJ tree.The results showed that there were few groups of different populations of Triplophysa pappenheimi clustering in a branch,indicating that the experiment selected five different groups of sample is relatively.(4)The comparative analysis results of the mitochondrial genome structure of Triplophysa pappenheimi and Triplophysa scleroptera,are as follows:Triplophysa pappenheimi and Triplophysa scleroptera sequences were 16572 bp and 16570 bp.T,C,A,G base content almost the same,the content of A+T were 56.5%,56.7%,there were AT bias,with vertebrate mitochondrial genome base composition.The length of the gene fragment of the coding protein is exactly the same.Triplophysa pappenheimi and Triplophysa scleroptera's 12 S rRNA gene'16S rRNA gene was located between tRNA-Phe and tRNA-Leu1,two kinds of Triplophysa 12 S rRNA gene fragment length are the same(951 bp),16 S rRNA gene of Triplophysa pappenheimi is 1678 bp,Triplophysa is 1679 bp;two kinds of Triplophysa contain 22 tRNA gene;control region(D-loop)was located between tRNA-Glu and tRNA-Phe genes,...
