With the development of sheep industry,multiple births sheep are favored.Hu sheep as one of the domestic breeds of multiple births sheep,It's lambing rate reach 229%[3].Pregnancy Toxemia of ewe commonly known as double lamb disease[5],more common in double lamb ewe or more than the ewe.At present,the study of toxemia at home and abroad mainly from nutritional metabolism and disease treatment,rare studies of the molecular level.In this text,266 Hu sheep as the research object were divided into experimental group(onset group,n=204)and control group(normal group,n=62),the lipid metabolism gene polymorphism was correlated with gestational ketoneemia and related indexes correlation analysis.This study aimed to find genetic markers associated with Pregnancy Toxemia of ewe studied these gene polymorphism and its correlation with Pregnancy Toxemia and its rich polymorphism gene in production,for the establishment of a new marker-assisted selection method provides the theory basis.(1)First,seven kinds of serum biochemical indexes(Glu?Ca?p?Mg?urea?AST?ALT),5 common physiological indicators and D-3-hydroxybutyric acid of sheep were detected.We known that the contents of Glu and Ca in the test group were significantly lower than those in the control group by single factor analysis of variance,(p<0.01);AST,ALT and D-3-hydroxybutyric acid were significantly higher than those in the control group(p<0.01);Test group D-3-hydroxybutyric acid concentration equal to 4.63mmol/L higher than the determination of toxins in pregnancy ? 3.0mml/L.In summary,the test group hypoglycemia,hypocalcemia,hyperglycemia,two liver function enzymes increased.(2)In this study,DNA mixed pooling method and Sequenom Mass Array with gene sequencing was used to analysis the genetic variation of HMGCS2 and DAGLA genes in sheep.A total of 17 mutation sites were found,8 synonymous mutation sites1 utr mutation site and 1 missense mutation site were found in DAGLA.4 intron variants 2 synonymous codons and one new Mutation site were found in HMGCS2.The chromosome number is Chr.1:96116630 of the new Mutation site.(3)Two genotypes in Fecb of Hu sheep were found in this study.It is BB and B+.The incidence of BB genotype was significantly higher than that of type B +(p<0.01);D-3-hydroxybutyric acid of BB type was significantly larger than B+ type(F=264.36,p=2.42E-36<0.01);(4)11 different haplotypes were found in GLAGA The frequency of haploid C-C-T-A-C-C-A-G-A in test group was significantly higher than that in control group(OR=2138731.13(2138730.22-2138732.04),p<0.001);It was found that ewes containing C-C-T-A-C-C-A-G-A haploid were more susceptible to toxemia.9different haplotypes were found in HMGCS2.The frequency of haploid T-A-C-T-T-G-C in the test group was significantly higher than that in the control group(OR=2.07,p=0.035<0.05);The frequency of haploid T-A-C-T-T-A-C in the test group was significantly higher than that in the control group(OR=394523444.81,p<0.0001);It was found that ewes containing T-A-C-T-T-G-C ? T-A-C-T-T-A-C haploid were more susceptible to toxemia.In conclusion,the polymorphism of Fecb gene was associated with the incidence of ketoprenalemia.GLAGA(C-C-T-A-C-C-A-G-A)and HMGCS2(T-A-C-T-T-G-C,T-A-C-T-T-A-C)were associated with the pathogenesis of toxemia in Hu sheep;Candidate genes for assisted breeding.
