Enolase(?-Enolase)expressed in the cytoplasm and nucleus as the key enzyme of glycolytic,which can convert 2-phosphoglycerate into phosphoenolpyruvate.The previous studies showed that ENO1 expression(P<0.05)in ovaries in laying goose,prompting us that ENO1 may promote follicle development and ovulation.This experiment studys the effect of ENO1 on the mRNA expression of follicle-stimulating hormone receptor(FSHR),estrogen receptor(ESR1),estrogen receptor(ESR2)and luteinizing hormone receptor(LHR)mRNA in primary cultured granulosa cells and the secretion of progesterone,estradiol and follistatin in the culture fluid for exploring the effect of ENO1 overexpression of avian granulose cells on receptor expression and secretory function.The study could providing scientific and theoretical basis for ENO1 function and laying performance of poultry.Five laying Zi geese 8 months were sacrificed by cervical dislocation post anesthesia.Granulose cells were obtained from the F1 follicles for 72 h.The test is divided into the culture group,Ad-CMV-Null group and Ad-CMV-ENO1 group,transfection condition is MOI=350 after48 h,collected cells and cell supernatant.And therefore,the research studys the effect of ENO1 on the mRNA expression of follicle-stimulating hormone receptor(FSHR),estrogen receptor(ESR1),estrogen receptor(ESR2)and luteinizing hormone receptor(LHR)m RNA in primary cultured granulosa cells and the secretion of progesterone ? estradiol and follistatin by the step double antibody sandwich enzymelinked immunosorbent assay(ELISA)in the culture fluid.The results are as follows:(1)The effect of overexpression of ENO1 on the level of the expression of mRNA of FSHR?ESR1?ESR2 and LHRLHR and ESR2 mRNA expression of the Ad-CMV-ENO1 group increased signigicantly comparing with the culture group and the Ad-CMV-Null group(P<0.01),but the expression mRNA of FSHR was significantly decreased(P<0.01),and the expression mRNA of ESR2 was no significant.(2)The effect of ENO1 overexpression of granulose cells on the secretion of progesterone?estradiol and follistatin in the culture fluidThe granulosa cells progesterone secretion of Ad-CMV-ENO1 group increased signigicantly comparing with the culture group and the Ad-CMV-Null group(P<0.01),however,the secretion of follistatin decreased signigicantly(P<0.01),and the secretion of estradiol was no significant(P>0.05).The results showed that the level overexpression of ENO1 affected the expression mRNA of FSHR?ESR1 and LHR significantly and the secretion of progesterone,follistatin,which suggested that ENO1 overexpression may affect these genes expression and orsteroid hormone secretion to improve granulose cells proliferation,therefore increase follicle development and ovulation.
