| The pathogens of gosling are goose parvovirus, goose and Muscovy duck at4-20day age can infected, and the other kinds of mammal could not be affected. The transmission of GPV are horizontal and vertical propagation, GPV spreads by alimentary canal called horizontal propagation, in addition GPV spreads by hatching eggs is vertical propagation, and also could be spread by embryo for vertical propagation. Gosling and recessive infected adult goose are the major infection source of goose parvovirus. The disease can occur yearly, but chiefly occur in spring and winter. The disease has the characteristics of fast spread, short course and high mortality, which seriously menace goose industry.To prepare high virus titer of the cell culture adapted goose parvovirus (GPV), the primary goose embryo fibroblast (GEF) was prepared and continuingly passed for31passages, and the YN isolate of GPV was adapted in the GEF for20passages. In addition, the virus titers were increased steadily from101.5TCID50/0.1mL of the initial virus (F0) to106.16TCID50/0.1mL of the cell adapted virus (F20) which shown the significant cytopathic effect in GEF after72hours post inoculation. Furthermore, the whole genomic sequences of the F0and the F20virus were sequenced, respectively, and the alignment analysis showed that the mutations were found133for nucleotides and22for amino acids between the two viruses. Of which the nucleotide mutations located in5’ and3’non-coding regions and the amino acids mutations located in VP3protein region, which could be the essential mutations for the virus adaption in GEF. |
