There are a great deal of barren hills and wasteland in Southern China.However,the quantity and the kinds of seedling supplied by the market of forest seedling are few.So how to product abundant seedling to improve and develop these barren hills and wasteland is an urgent issue.In view of this,this paper took the Jing county No.86005 clone,which was better,as research material based on amount of research of screening for Chinese Fir superior clones which were suitable for planting in the hilly region.The study main contents explants chosen,the establishment of the best sterilization system,and searching of the best medium and other conditions for induction culture,multiplication culture and rooting medium.The main results are as follows:(1)The best system of sterilization:First clear the detergent solution on the materials.Then the stem was soaked in 75%alcohol for 30 seconds,and followed 11-minutes’ treatment of 0.1%HgCl2.By this method,the survival rate was nearly 100%.When pre-treating the shoot tips,the leaves should be kept.And the sterilization system,soaked in 75%alcohol for 30 seconds,then 7-minutes’ treatment of 0.1%HgCl2,worked well for shoot tips.By this method,the survival rate was 99.24%.(2)The best medium for stem induction culture was MS + 0.8mg/L 6-BA +0.4mg/IBA + 6 g/L Agar + 30 g/L Sucrose.After 45 days,the effective induction rate was 95.83%and the average number of buds was 6.3.(3)It’s the best time for multiplication culture when the length of bud at 1.8-2.2 centimeters.MS +0.1 mg/L IBA + 0.3 mg/L 6-BA+6 g/L Agar+30 g/L Sucrose has the best effect on the multiplication culture.And the height growth of bud was 1.71 centimeters after culturing 20 days,the multiplication coefficient was 3.4 after 45-days’ culturing.(4)The optimum rooting medium was 1/2MS + 0.1 mg/L NAA + 0.8 mg/L IBA+6 g/L Agar+30 g/L Sucrose,and the rooting rate was 82.33%.
