The tissue culture of Chinese jujube cultivar "Dongzao"widely cultivated in China andwild jujube was studied. Main results were as follows:1. Study on sterilization of explent of tissue culture in Chinese jujube.Compared effects of different sterilizations on explants,the results(include contaminationrate, browning rate, survival rate) showed that at the beginning of growing period, the beststerilization method was that explants could be treated with 0.1ï¼…HgCl2 for 6 minutes. Duringfast-growing period, the contamination rate of explants was decreased by 0.1ï¼…HgCl2 whichtreated explants twice, and every time for 6 minutes. The new shoots from dormancy branchescultured in water could be treated by 0.1ï¼…HgCl2 for 4 minutes. There are no resolvent forendogenesis contamination.2. Study on sampling of explants around the year in the tissue culture of Chinese jujube.In order to prolong the period of sampling explants in tissue culture, the explants weresampled every month, and coming from different places and positions. The results showedthat explants in tissue culture of Chinese jujube could be sampled around the year, and thebest sampling time between November and next June. The first head-branch is better thanother types of branches.3. Study on rapid propagation mediums of Chinese jujube.Suitable conditions for tissue culture and rapid propagation of"Dongzao"and wild jujubewere studied using their branches as material. The results showed that for "Dongzao" thebasic MS medium supplemented with 2.0mg/L 6-BA, 0.2mg/L IBA was the suitable initiationmedium and MS+6-BA(1.0mg/L)+IBA(0.1mg/L) was the best proliferous medium. For wildjujube, the optimized initial and proliferous medium were MS+6-BA(1.0mg/L)+IBA(0.1mg/L)andMS+6-BA(2.0mg/L) separately.
