In this paper, rhizosphere soil of Angelica sinensis- soybean rotation and A. sinensis planting in Huichuan, Weiyuan County of Gansu Province were regarded as research materials, the soil of surrounding idle was regarded as the comparison of experiments. By constructing ITS colony library, sequencing and the analysis of restriction enzyme map according to three king of soil fungi from experimental soil;then, the diversity and composition of the population were compared; the vary of fungi number and construction from the A. Sinensis rhizosphere soil under the different planting styles were studied; through discussing the mechanisation of succession cropping obstacle, which provided the theoretical basis for better A.sinensis rotation.The results of this study were as follows:(1) Constructing ITS colone library and analysis of restriction enzyme mapsBy constructing ITS clone library and analysis of restriction enzyme maps, we concluded that the restriction enzyme clusters of the fungi ITS clone library from three sample soils had significant differences and the clone number were also different.(2)diversity analysis of fungi population from three kinds of sample soilThe Shannon Diversity Index and Schaol Index of the soil fungi: A. Sinensis planted fields ?rotation planted fields?uncultivated land, which indicated that fungi population was more than A. sinensis-soybean rotation planted fields. The Simpson Index and Species Evenness Index were as same as the above, which showed that the diversity of planting style would lead to different influence on distribution pattern and dominance degree of fungi; fungi dominance degree of A. sinensis planted field was significant and the distribution of fungi population was uniformly. The different planting styles would result in the diversity vary of soil fungi from A. sinensis rhizosphere.(3) The analysis of fungi population construction from three sample soil In this study, we obtained 24 genera from 6 groups according to ITS clone library of fungi from three sample soil. in A. sinensis planted field, Fusarium, Colletotrichumand Alternaria were important dominant groups. Cheatomium globosum and Mortierella were important dominant groups in A. sinensis-soybean rotation land;however, the dominant groups were not significant in wasted land. Phytopathogenic fungi, for example Alternaria, Rhizopus et al. was only found in A. sinensis rotation land; the above states indicated that the accumulation of pathogens was one of reasons leading to succession planted obstacle. Different planting methods caused the variety of fungi in angelica rhizosphere soil.
