Over thousand of Galba pervia were collected from 40 different regions of Guangxi to survey the fluke infection in the intermediate host.G.pervia was crushed to test the infection of fluke and identified larvaes species using fluke universal primers.The infected G.pervia species were confirmed by specific primers as well.Results showed that a total of 12 different regions detected positive snails.All the positive snails identified as G.pervia with the specific primers.Echinostoma revolutum,Alaria alata and Plagiorchis elegans were identification in snail with universal primers and sequencing.Echinostoma revolutum is the most common fluke in G.pervia and appeared in 11 regions.The results concluded that G.pervia of Guangxi could be used as an intermediate host for a variety of flukes in Guangxi.In order to preliminarily understand the internal defense of G.pervia system.The slices of G.pervia lymphocytes were making and observed the morphology and classification of lymphocytes.Meanwhlie,it's to observe the pathological changed of G pervia between uninfected and infected with Fasciola gigantica.The results show that the lymphocytes of G.pervia were divided into three categories of stretch amoeboeyte,hyalinocyte,round amoeboeyte that the average diameter of 0.751um,0.672um,0.608um and the percent of the three cells are 47.79%,6.33%and 45.88%,respectively.Their digestive system is divided into the digestive tract with mucous membrance,submucosa,lamina muscularis and adventitia and digestive gland with columnar cells and goblet cells.The lymphocytes of G.pervia infected with Fasciola gigantica were to increase in the parasitic parts.The pathological phenomena of liver were that secretions increased,goblet cells into the vacuole,glandular epithelium loosely off,structure of the entire liver fuzzied.Shows:lymphocytes of G pervia may play a role of defense when foreign invasion.To understand the anatomic and phylogenetic of G pervia from Guangxi,G pervia from the same place were analysis the conchology and anatomical and the principal component.Meanwhile,the sequence of the mitochondrial coxl gene of G pervia from 18 regions were amplification and sequencing.The phylogenetic tree based on the coxl gene was constructed.Results:the principal component analysis showed that the majority of G pervia were together and the other was a discrete distribution,so that the intraspecific of G pervia were some dififerences.The phylogenetic tree of coxl gene in G pervia could be divided into four clades(?,?,?,?)and 69%of the G pervia sequences belong to clade ?.The same geographical sources of the snails did not have a strong relation with the phylogenetic tree analysis.Shows:G pervia may be have different genotypes.
