This study breeding Large spike lines 1-5?1-6?1-7?1-8?1-9?1-10?1-11 from the Hybrids of white-rye Substitution lines 5R/5A and 1R/1D,we breeding lines 1-12?1-13?14?1-15?1-16?1-17?1-18?1-19?1-20?1-21?1-22?1-23?1-24 from the hybrids of white-rye Substitution lines 6R/6A and 1R/1B,we detect these 20 advanced lines by morphology?cytology and molecular markers,to get the Breeding base material of R group,The main results obtained are as follows:1.the result of Morphological and cytological show that the genetic performance of 20 lines were stability,and have the good rye traits,such as big spike,resistance to disease.We can breeding the plants can be observed directly have both parents traits from 20 lines,such as spikelets of substitution lines of 1R is arranged in neat rows,resistance to stripe rust,leaf rust,stem rust;substitution lines of 5R have the long spike,stem hair,ear sparse,etc.substitution lines of 6R is wide ear,awnless,high resistance to Powdery Mildew,etc.the Observation results of Root tip Chromosome show that it have 42 Root tip Chromosome,The meiosis Metaphase I chromosome configuration of Pollen mother cells is stably,Observations result show that the experimental materials is basically stable on cytology.2.the results of Molecular marker test show that 20 lines are Imported rye chromosome that is confirmed by rye-specific primers PSC119.1.The amplification results of primer SCM138 showed that 1-6,1-7,1-8,1-9,1-10 are imported rye chromosome 5RS fragment,primer SCM120 can amplify specific fragment of rye 5R long arm stably in 1-5?1-6?1-7?1-8?1-9?1-10?1-11,primer TSM604 can amplify specific fragment of rye 1R short arm stably in 15 lines,primer SCM180 can amplify specific fragment of rye 6R long arm stably in 1-15?1-17?1-18?1-20?1-21?1-22?1-23,To victory rye as probe,We performed in situ hybridization on it,Proximal part containing a rye chromosome appears The hybridization signal,fragment is small,we Judge for the small fragment translocation lines.These results provide the material foundation for wheat genetic breeding and quality improvement.
