| Chelidonium majus L.belongs to the family Papaveraceae and is a rich source of various biologically active substances such as alkaloids,and it has anti-inflammatory,anti-fungal,anti-bacterial,anti-tumor and other biological activity.The contents of alkaloids has a large difference between different harvest period,different place of origins and parts,and the type and content of alkaloid components could be used as the internal factors to judge the quality of C.majus.In this study,we mainly proceeded from the secondary metabolites of C.majus to track the change of alkaloid content in one year and obtain the peak period of alkaloid content,and determined the best harvest period.In addition,we also determined the differences in alkaloid contents of C.majus from different origin,and analyzed the key factors that affected the content of alkaloids.At the same time,this study measured the content of the primary metabolites such as soluble sugar,soluble protein and water in the wild C.majus L from 1 year,and tried to explore the correlation between primary metabolism and secondary metabolism.we used transcriptome sequencing technology to reveal the biosynthesis mechanism of alkaloids,and focused on the genes involved in the synthesis pathway,and found the correlation between gene expression and the content of alkaloids in different growth stages,and provide a reliable theoretical basis for domestication and cultivation of C.majus.The main results were as follows:(1)Determination of six alkaloids in the C.majus,include protopine,chelidonine,berberine,coptisine,sanguinarine and chelerythrine,by using the high performance liquid chromatography(HPLC).It is considered that the content of each component in the end of the end of June and the end of July is high and the biological yield is large,which can be used as the best harvest period.(2)The differences in the content of alkaloids in C.majus from Qingyuan County of Liaoning Province,Ji’an City of Jilin Province,Yichun City of Heilongjiang Province and Genhe city of Inner Mongolia roots and rivers in the same growth and development period were measured.The results show that the higher latitude are,the higher the alkaloid content,and the plants are large.(3)The water content,soluble sugar content and soluble protein content of wild C.majus L in the year were measured.It was found that the water content of C.majus was gradually increased from the dormancy period to growing period,and the soluble sugar and soluble protein were gradually decreased.(4)The differential genes from five period in the synthesis pathway of isoquinoline alkaloids were analyzed by transcriptome sequencing technique,and chosen two genes which encoding the key(5)enzymes :(1)(S)-coclaurine N-methyltransferase(CNMT)and aspartate aminotransferase(GOT2),and the expression of these two genes was show that some correlation in the sanguinarine and chelerythrine synthesis. |
